脑肠肽激素Ghrelin对胰岛素抵抗骨骼肌细胞质游离钙离子及葡萄糖转运子4影响的研究

摘　　要：目的探讨IR的骨骼肌细胞质游离钙离子(Ca2+)的变化,脑肠肽激素Ghrelin对Ca2+稳态的维持作用及其引起细胞内葡萄糖转运子4(GluT-4)的变化。方法建立IR的骨骼肌细胞模型,将实验细胞分为单纯胰岛素对照组(Con)、棕榈酸处理组(PA)、酰化Ghrelin处理组(AG)、非酰化Ghrelin处理组(UG)及人肥胖抑制素(Obestatin)处理组(OB)。采用GOD法、流式细胞仪、激光共聚焦显微镜检测Ghrelin对棕榈酸诱导的IR的缓解作用、细胞质游离Ca2+及GluT-4的水平变化。结果与PA组比较,AG、UG、OB组培养基细胞上清液葡萄糖浓度降低(P<0.05)。与PA组比较,AG、UG、OB组细胞质游离Ca2+及GluT-4水平升高(P<0.05),且变化趋势一致。结论 IR形成后细胞质游离Ca2+水平降低,经Ghrelin处理后可改善IR,细胞质游离Ca2+水平升高,同时加强细胞内GluT-4的生理作用。Objective To investigate the changes of calciumion(Ca2+)in skeletal muscle cytoplasm of insulin resistance(IR),the effect of Ghrelin on Ca2+homeostasis and the changes of intracellular glucose transporter 4(GluT-4).Methods A skeletal muscle cell model of IR was established.The experimental cells were divided into a simple insulin control group(Con),a palmitate treatment group(PA),an acylated Ghrelin treatment group(AG),a non-acylated Ghrelin treatment group(UG),and an obesity inhibin(Obestatin)treatment group(OB).The glucose oxidase method,flow cytometry,and laser confocal microscopy were used to detect the alleviating effect of Ghrelin on palmitate-induced IR,changes in cytoplasmic free Ca2+levels,and changes in GluT-4 levels.Results The glucose concentration in the cell supernatant was reduced in the AG,UG,and OB groups compared with PA group(P<0.05).Intracellular Ca2+and intracellular GluT-4 levels increased in the AG,UG,and OB groups than in PA group(P<0.05),which Conclusion After the formation of IR,the intracellular free Ca2+level was reduced.After Ghrelin treatment,IR could be improved,the intracellular free Ca2+level was increased,and the physiological effects of GluT-4 were enhanced in the cell.
