摘 要:目的:通过研究解毒祛瘀滋肾方对mBAFF刺激后MRL/lpr狼疮小鼠BAFF/BAFF-R信号通路的影响,探讨解毒祛瘀滋肾方治疗系统性红斑狼疮增效减毒的机制。方法:将MR L/l pr狼疮小鼠随机分为4组:空白组(C)、mBAFF刺激组(R)、mBAFF+强的松组(G)、mBAFF+解毒袪瘀滋肾方组(J),Balb/c小鼠设为正常对照组。干预8周后,计算各组小鼠脾脏、胸腺指数,RT-PCR实验检测各组小鼠脾脏和胸腺BAFF、BAFF-R、NF-κB、Bcl-2、IL-10 mRNA的表达,流式细胞术检测各组小鼠脾脏淋巴细胞的凋亡。结果:在mBAFF刺激之后,MRL/lpr狼疮小鼠脾脏、胸腺指数以及脾脏、胸腺的BAFF、BAFF-R、NF-κB、Bcl-2、IL-10 mRNA的表达水平均显著上调(P<0.05),同时MRL/lpr狼疮小鼠脾脏淋巴细胞的凋亡率也随之下降;解毒祛瘀滋肾方和GC分别干预后,MRL/lpr狼疮小鼠脾脏及胸腺指数均有显著下降(P<0.05),脾脏淋巴细胞凋亡率也伴随着BAFF、BAFF-R、NF-κB、Bcl-2、IL-10 mRNA表达的降低而升高。结论:解毒祛瘀滋肾方治疗SLE增效减毒的机制可能是通过调控异常活化的BAFF/BAFF-R信号通路来实现的。Objective:To study the effects of Jiedu Quyu Zishen Formula on the BAFF/BAFF-R signaling pathway of MRL/lpr lupus mice stimulated by mBAFF,and to explore the mechanism of Jiedu Quyu Zishen Formula in the treatment of systemic lupus erythematosus(SLE).Methods:MRL/lpr lupus mice were randomly divided into 4 groups:control group(C),mBAFF stimulation group(R),mBAFF+GC group(G),mBAFF+Jiedu Quyu Zishen Formula group(J),and Balb/c mice were set as the control group.After 8 weeks of intervention,the indexes of spleen and thymus were calculated.The mRNA expressions of BAFF,BAFF-R,NF-κB,Bcl-2 and IL-10 were detected by RT-PCR.Flow cytometry was used to detect the apoptosis of spleen lymphocytes in mice.Results:After mBAFF stimulation,the expression levels of BAFF,BAFF-R,NF-κB,Bcl-2,IL-10 mRNA in spleenand thymus index and spleen lymphocytes of MRL/lpr lupus mice were significantly up-regulated(P<0.05),while the apoptotic rate of spleen lymphocytes of MRL/lpr lupus mice were also decreased(P<0.05).However,the spleen and thymus index of MRL/lpr lupus mice decreased significantly after intervention with Jiedu Quyu Zishen Formula and GC respectively,and the apoptosis rate of spleen lymphocytes is also increased with the decrease of BAFF,BAFF-R,NF-κB,Bcl-2 and IL-10 mRNA expression.Conclusion:The mechanism of detoxifying,removing blood stasis and nourishing kidney prescription in the treatment of SLE may be realized by regulating abnormally activated BAFF/BAFF-R signaling pathway.
