摘  要:目的:考察生物相容性党参远志散微乳对阿尔茨海默病(AD)模型小鼠学习记忆的影响,并从抗氧化角度分析其作用机制。方法:ICR小鼠60只随机分为空白组、模型组、空白微乳组、阳性药组、党参远志散水提组及微乳组。除空白组外,其余组均腹腔注射D-半乳糖和亚硝酸钠建立复合AD模型,分别灌胃蒸馏水、空白微乳、脑复康、党参远志散水提液及生物相容性党参远志散微乳6周。采用Step-down实验和Morris水迷宫评价学习记忆能力,生化方法检测脑组织中SOD、CAT活力及MDA含量,HE染色法观察海马组织病理形态学改变。结果:与模型组比较,党参远志散微乳组的学习记忆能力显著提高(P<0.05,P<0.01),脑组织中SOD、CAT活力显著升高(P<0.01),MDA含量显著降低(P<0.01),海马组织的病理结构改变得到改善;空白微乳组脑组织中SOD活力显著升高(P<0.01)。结论:生物相容性党参远志散微乳可通过氧化应激提高AD模型小鼠学习记忆能力,空白微乳在AD治疗中起到一定辅助作用。Objective:To investigate the effects of biocompatible microemulsion extract of compound Dangshen Yuanzhi on learning and memory abilities in mice model with AD and analyze the mechanism from anti-oxidation function.Methods:Sixty mice were randomly divided into normal group,model group,biocompatible microemulsion group,positive drug group,water extract group and microemulsion extract group.AD model were established by intraperitoneal injection with D-galactose and sodium nitrite.Meanwhile,each group was given by gavage for 6w with distilled water,biocompatible microemulsion,water extract and microemulsion extract.The learning and memory abilities were evaluated by Step-down test and Morris water maze.Activities of SOD and CAT,contents of MDA in brain tissue were measured by biocnemical methods.The pathological changes of hippocampal were observed by HE staining.Results:Compared with model group,learning and memory abilities of microemulsion extract group were significantly improved(P<0.05,P<0.01),activities of SOD and CAT were significantly improved(P<0.01),contents of MDA were significantly decreased(P<0.01),the pathological changes of hippocampal in microemulsion extract group were improved activities of SOD of blank microemulsion group were significantly improved(P<0.01).Conclusion:Biocompatible microemulsion extract of compound Dangshen Yuanzhi Powder can improve the learning and memory abilities of AD mice significantly through oxidative stress,biocompatible microemulsion can be an assistant role in the treatment of AD.
