摘 要:目的:研究时相调节对方对常用光环境下兔眼轴的影响,探讨其对视网膜多巴胺(DA)及其D1受体的作用与机制。方法:将1月龄新西兰白兔24只随机分为自然光组、自然光食疗组、荧光灯对照组和荧光灯食疗组,每组6只。运用时相调节对方对食疗组进行干预12周,对照组兔常规饲养。实验开始后每隔2周对各组兔眼进行眼轴的测量。并于第12周时,采用HPLC测定视网膜DA及Western Blot测定D1受体的相对蛋白含量。结果:各组幼兔眼轴较实验前均有增长,且在实验第2周起,荧光灯组眼轴长度大于自然光组(P<0.05,P<0.01);时相调节对方干预后,在实验第6周时,自然光食疗组幼兔眼轴长度比自然光组短(P<0.05);荧光灯食疗组幼兔眼轴长度均小于荧光灯组,并在第2、4、8、10周时差异有统计学意义(P<0.05)。视网膜DA含量在时相调节对方干预后,食疗组较对照组均有增加(P<0.05,P<0.01),且D1受体蛋白水平均小于对照组,与自然光组比较,自然光食疗组显著减少(P<0.01)。结论:时相调节对方能够通过调整阴阳平衡而发挥屈光保护作用,有效的抑制幼兔眼轴的发展,其作用机制可能跟抑制视网膜D1受体的水平,提高视网膜DA的表达有关。Objective:To study the effects of traditional Chinese medicine on the rabbits’axial length in common light environment,and to explore the effects on the dopamine and its D1 receptor in the retina.Methods:A total of 24 New Zealand white rabbits were randomly divided into 4 groups,nature light group,nature light diet group,fluorescent group,fluorescent diet group,6 for each group.Control group was exposed in natural light and fluorescent group was exposed in fluorescent light for 12 weeks,on which basis natural diet group and fluorescent diet group were fed with Shixiang Yinyang Formulas,respectively.The eye axis of each group of rabbit eyes was measured every 2 weeks after the experiment began.At the 12th week,the relative protein content of D1 receptor was determined by HPLC and Western Blot.Results:The axial length of all groups increased.At the 2nd week of the experiment,the axial length of fluorescent group was longer than that of natural light group(P<0.05,P<0.01).After the intervention of Shixiang Yinyang Formulas,at the sixth week of the experiment,the axial length of young rabbits in the natural light diet group is shorter than that in the natural light group(P<0.05).At the 2nd,4th,8th,and 10th week,the axial length of rabbits in the fluorescent diet group was shorter than fluoressent group(P<0.05).After 12 weeks,fluorescent diet group and natural diet group were increased in retinal DA levels(P<0.05,P<0.01),and decreased in the levels of retinal D1 receptors,and compared with the natural light group,the natural light diet group was significantly reduced(P<0.01).Conclusion:Shixiang Yinyang Formulas can exert refractive protection by adjusting the balance of yin and yang,and effectively inhibit the development of the axial length of rabbits.The mechanism may be related to inhibit the level of D1 receptors and increase the expression of retinal dopamine.
