唾液酸抗炎作用研究

摘　　要：目的:研究唾液酸的抗炎作用。方法:将健康SPF级成年雄鼠96只,随机分为空白组、唾液酸低、中、高剂量组(每组24只)。空白组给予超纯水,唾液酸3个干预组分别予以15、30、60mg/kg的唾液酸溶液灌胃30d。采用二甲苯致小鼠耳肿胀法(每组12只)和角叉菜胶致小鼠足跖肿胀模型(每组12只)观察唾液酸的抗炎作用,并测定血清中的CRP、TNF-α、IL-1β、IL-6水平和肿胀足部位前列腺素E2(PGE2)水平。结果:唾液酸干预能显著减轻角叉菜胶致小鼠足跖肿胀,与空白组比较,唾液酸中剂量组在注射角叉菜胶4、5、6h后足跖肿胀度显著降低(P<0.05),唾液酸高剂量组在4、5h后足跖肿胀度显著降低(P<0.05);唾液酸中剂量组小鼠血清中IL-6、TNF-α水平显著低于空白组(P<0.05),唾液酸低、中、高剂量组肿胀足PGE2水平均显著高于空白组(P<0.01)。结论:唾液酸具有显著的抗炎作用,其中30mg/kg剂量组抗炎效果较好。Objective:To investigate the anti-inflammatory effect of sialic acid,and explore its possible mechanism.Methods:A total of 96 healthy adult SPF male mice were randomly divided into 4 groups(24 mice in each group),including control group and low sialic acid,medium and high intervention group.The blank group was administered with ultrapure water,and the three sialic acid intervention groups were administered with 15,30,and 60mg/kg of sialic acid solution for 30 days.Xylene induced ear edema experiment(12 mice in each group),and carrageenan induced foot swelling experiment(12 mice in each group)were adopted to evaluate the anti-inflammatory efficacy of sialic acid.Meanwhile,levels of CRP,TNF-α,IL-1βand IL-6 in serum and PGE2 in the swollen feet were determined.Results:Sialic acid intervention can significantly reduce carrageenan-induced swelling of mice.Compared with the blank group,the sialic acid medium-dose group significantly reduced the swelling of mice at 4,5,and 6 hours after carrageenan injection(P<0.05).Foot swelling of mice in the high-dose sialic acid group significantly decreased at 4 and 5 hours(P<0.05).Serum levels of IL-6 and TNF-αin mice treated with the medium-dose sialic acid intervention group were lower than those in the blank group(P<0.05).The swollen foot PGE2 level in the low,medium and high sialic acid intervention group was significantly higher than that in the blank group(P<0.01).Conclusion:Sialic acid has obvious anti-inflammatory effects,and the anti-inflammatory effect of the 30mg/kg dose group is better.
