肾衰方及其拆方对单侧输尿管梗阻大鼠肾组织PAI-1、E-cadherin表达的影响

摘　　要：目的:观察中药肾衰方对大鼠肾间质纤维化肾组织PAI-1、E-cadherin表达的调节及其抗肾间质纤维化的作用机制。方法:将72只大鼠随机分为:假手术组、模型组、肾衰方组、祛邪方组、补虚方组、贝那普利组,采用单侧输尿管梗阻(UUO)动物模型,术后第7、14天分别观察梗阻侧大鼠肾组织病理改变,免疫组化检测肾组织PAI-1、E-cadherin的表达。结果:术后第7、14天,模型组与假手术比较大鼠肾组织E-cadherin表达下降(P<0.05),PAI-1表达增强(P<0.05),而经肾衰方、祛邪方、补虚方和贝那普利治疗后较模型组大鼠肾组织E-cadherin表达提高(P<0.05),PAI-1表达下降(P<0.05);术后第7、14天肾衰方组优于祛邪方组、补虚方组、贝那普利组(P<0.05)。结论:肾衰方及其拆方可能是通过诱导E-cadherin表达,抑制PAI-1表达,减轻肾间质的纤维化,并通过以方测证,说明肾间质纤维化为本虚标实之证。Objective:To observe the effect of the traditional Chinese medicine of Shenshuai Formula on the expression of PAI-1 and E-cadherin in rat renal interstitial fibrosis and the mechanism of its anti renal interstitial fibrosis.Methods:Seventy two rats were randomly divided into sham operation group,model group,Shenshuai Formula group,the Quxie Formula group,Buxu Formula group and the Benazepril group.The animal model of unilateral ureteral obstruction(UUO)was adopted and the pathological changes of renal tissue in the obstructed rats were observed on the 7th and 14th days after surgery,and the expressions of PAI-1 and E-cadherin in the renal tissue were detected by immunohistochemistry.Results:On the 7th and 14th day after operation,the expression of E-cadherin in the renal tissue of the model group decreased compared with that of the sham operation(P<0.05),and the expression of PAI-1 increased(P<0.05).On the 7th and14th day after operation,Shenshaui Formula group was superior to the Quxie Formula group,the Buxu Formula group and the Benazepril group(P<0.05).It is indicated that renal interstitial fibrosis belongs to the syndrome of deficiency in origin and excess in superficiality.Conclusion:Shenshuai Formula and its components may induce the expression of E-cadherin,inhibit the expression of PAI-1,and alleviate the fibrosis of renal interstitial,and the renal interstitial fibrosis is the syndrome of deficiency in origin and excess in superficiality.
