摘 要:为了解安徽省部分地区散养鸡球虫的分布现状与感染类型,2018年5-6月间从亳州市谯城区、蚌埠市固镇县、合肥市肥东县、安庆市潜山县和宣城市宣州区5个行政区11个村落中随机采得散养鸡新鲜粪样1000份并提取DNA;合成8对特异性引物,分别对鸡球虫ITS1基因及各虫种特异性片段进行PCR扩增和测序分析。结果显示:共检出鸡球虫阳性样品111份,总感染率为11.10%,所有艾美耳球虫公认种均被检出,未见5种及以上虫株共感染现象;肥东散养鸡感染率最高,感染类型最复杂,共检出5种艾美耳球虫,是4种球虫共感染类型和柔嫩艾美耳球虫的唯一检出地;谯城区感染率为8.44%(19/225),除未检到柔嫩艾美耳球虫外,其他虫株阳性检出均与肥东相同;固镇散养鸡感染率为6.57%(14/213),单一虫种感染为常见类型,堆型艾美耳球虫与和缓艾美耳球虫仅在此地检出;宣州鸡球虫感染率为3.19%(6/188),检出3种艾美耳球虫,除毒害艾美耳球虫外其他虫株与亳州谯城相同;潜山球虫感染率最低(1.00%,2/200),仅检出布氏艾美耳球虫和毒害艾美耳球虫。除蚌埠固镇与亳州谯城、宣城宣州间,及宣城宣州与安庆潜山间无统计学意义外,其他各调查点间鸡球虫的总感染率均差异显著(P<0.05)。结果表明,安徽省散养鸡群的球虫多态性丰富,不同地区总体感染率间差异明显,混合感染类型较为常见,应当成为鸡球虫病流行的防控重点。To investigate the prevalence of coccidiosis in free-range chickens in Anhui Province and to identify the type of this infection,1000 fresh feces samples were randomly collected from the free-range chickens in eleven villages of five administrative regions in the Anhui area during the period of May to June in 2018 for DNA extraction.The sampled regions included Qiaocheng District of Bozhou City,Guzhen County of Bengbu City,Feidong County of Hefei City,Qianshan County of Anqing City,and Xuanzhou District of Xuancheng City.Eight pairs of primers were synthesized according to the declaratory sequences in the Gen Bank database and the published articles in order to evaluate the infection rates in the obtained samples and the mixed species types they belong to by PCR amplification of the ITS1 gene or the specific fragments of the Eimeria species.The results indicated that 111 positive cases of coccidiosis including all the recognized Eimeria species were detected at a total infection rate of 11.10%,but no co-infection with five or more species was observed.The highest infection rate of coccidiosis of complex mixed species infection type was examined in Feidong County,in which five kinds of Eimeria including the E.tenella,and the co-infection with four Eimeria strains were detected.The infection rate in Qiaocheng District was 8.44%(19/225),and the same species as that in Feidong County were detected,except E.tenella.For free-range chickens in Guzhen County,the infection rate was 6.57%(14/213),and the infection with single species was common,including E.acervulina and E.mitis.The infection rate of coccidiosis in Xuanzhou District was 3.19%(6/188),and the same Eimeria species as those in Qiaocheng District were detected,except E.necatrix.The lowest infection rate(1.00%,2/200)without any mixed species infection was detected in Qianshan County.Notably,the total infection rate of coccidiosis was statistically different among all the survey points except those of Guzhen vs Qiaocheng,Guzhen vs Xuanzhou,and Xuanzhou vs Qiansha
