摘 要:目的:探讨在超高温饲养环境下,芍药甘草汤及其拆方抗高温环境大鼠死亡机理与血清乙酰胆碱(Ach)、前列腺素E2(PGE2)和脑、心、肝组织热休克蛋白70(HSP70)含量变化的关系。方法:实验将SPF级SD大鼠饲养于超高温(36℃~39℃)环境中,记录大鼠死亡率,于给药13d后,采集血清检测Ach、PGE2含量、取脑、心、肝组织检测HSP70含量。结果:与高温正常组相比,芍药甘草汤、单味白芍、单味甘草组大鼠死亡率均有不同程度的下降,死亡率分别为33.33%、46.67%和26.67%;超高温环境可使大鼠血清Ach、PGE2和脑、心、肝组织HSP70含量均呈上升趋势,其中以血清Ach和脑、肝组织HSP70含量上升幅度尤为显著(P<0.05,P<0.01);各给药组均可显著对抗高温所致大鼠脑组织HSP70含量的增加(P<0.05,P<0.01),而芍药甘草汤组和单味白芍组则可协同增加高温所致大鼠血清PGE2的含量。结论:超高温环境大鼠的死亡机制与血清Ach、PGE2和脑、心、肝组织HSP70含量的变化密切相关,而芍药甘草汤及拆方耐高温机理可能与上调血清PGE2水平进而干预大鼠脑、心、肝组织HSP70活性有关。Objective:To explore the relationship between the death mechanism of rats in high temperature with Shaoyao Gancao Decotion and its disassembled prescription and the content changes of acetylcholine(Ach)and prostaglandin E2(PGE2)in serum,and heat shock protein 70(HSP70)in brains,hearts and livers of rats under ultra-high-temperature rearing environment.Methods:Specific patho-gen free(SPF)grade SD rats were fed in ultra-high temperature environment(36℃~39℃)in the test.The mortality of rats was recorded.After 13 days of treatment,the content of Ach and PGE2 in serum and HSP70 in brain,heart and liver were detected.Results:Compared with the hyperthermic normal group,the mortality of rats in Shaoyao Glycyrrhiza Decoction group,Paeonia lactiflora group and Glycyrrhiza uralensis Fisch group decreased in varying degrees,which were 33.33%,46.67%and 26.67%,respectively.The contents of Ach,PGE2 in serum and HSP70 in brain,heart and liver of rats,especially Ach in serum and HSP70 in brain and liver(P<0.05,P<0.01),increased in ultra-high temperature environment.Treatment groups could significantly increase the HSP70 content in brain induced by high temperature(P<0.05,P<0.01),while Shaoyao Glycyrrhiza Decoction group and Paeonia lactiflora group could jointly increase the content of PGE2 in serum of rats induced by high temperature.Conclusions:The death mechanism of rats in ultra-high temperature environment is closely related to the content changes of Ach,PGE2 in serum and HSP70 in brain,heart and liver.The mechanism of high temperature tolerance of Shaoyao Gancao Decoction and its disassembled prescription may be related to the up-regulation of PGE2 level in serum and and intervention of HSP70 activity in brains,hearts and livers of rats.
