摘 要:目的观察莱菔子对胃肠积热大鼠胃肠动力的影响,并探讨其作用机制。方法选择60只SPF级雄性大鼠,随机分为正常组、胃肠积热组及莱菔子低、中、高、超高(0.015、0.03、0.06、0.12 g/mL)剂量组。正常组给予普通饲料喂养,胃肠积热组、莱菔子各剂量组给予高热量饲料喂养及52%牛奶溶液,莱菔子各剂量组给予莱菔子混悬液灌胃干预。观察各组大鼠体征,取大鼠胃及结肠组织做HE染色,行小肠推进实验,用ELISA法检测血清中胃饥饿素(Ghrelin)、胃泌素(GAS)、P物质(SP)、一氧化氮(NO)的含量。结果胃肠积热组及莱菔子各剂量组大鼠体质量明显低于正常组(P<0.05)。胃肠积热组、莱菔子低剂量组、中剂量组小肠推进率明显低于正常组(P<0.05),超高剂量组小肠推进率高于胃肠积热组(P<0.05)。胃肠积热组、莱菔子低剂量组、中剂量组血清中Ghrelin含量明显低于正常组及超高剂量组(P<0.05)。胃肠积热组血清中NO含量高于正常组、莱菔子高剂量组、超高剂量组(P<0.05);胃肠积热组、莱菔子低剂量组血清SP含量明显低于正常组(P<0.05),而超高剂量组血清NO含量则明显高于胃肠积热组(P<0.05)。各组大鼠血清中GAS含量未见明显差异(P>0.05)。结论莱菔子可以升高胃肠积热大鼠血清中Ghrelin、SP含量,降低NO含量,增强小肠推进作用,从而促进胃肠积热大鼠的胃肠动力,但其作用与剂量有关,大剂量莱菔子的推动作用更加明显。Objective:To study the effects and the mechanism of radish seed on gastrointestinal motility in rats with gastrointestinal heat retention(GHR).Methods:60 SPF grade male rats were randomly divided into the normal group,GHR group and the dose groups of radish seed(low,medium,high and ultra high).The normal group was given ordinary feed;GHR group and radish seed groups was given high-calorie feed and 52%milk solution.Every dose group of radish seed was administrated with radish seed suspension.The physical signs of rats in each group were collected;the gastrointestinal tissues of rats were taken for HE staining,and the small intestine propulsion experiment was carried out.The contents of Ghrelin,GAS,SP and NO in the serum were detected by ELISA.Results:The body mass of rats in GHR group and dose groups were significantly lower than that in the normal group(P<0.05).The rate of small intestine propulsion was significantly lower in GHR group,low dose group and medium dose group than that in the normal group(P<0.05),and higher in ultra high dose group than that in GHR group(P<0.05).The levels of Ghrelin in the serum of GHR group,low dose group and medium dose group were significantly lower than those of the normal group and ultra high dose group(P<0.05).No content in serum of GHR group was higher than that of the normal group,high dose group and ultra high dose group(P<0.05).The serum SP contents of GHR group and low dose group were significantly lower than that of the normal group(P<0.05),while that of ultra high dose group were significantly higher than those of GHR group(P<0.05).There were no significant differences in the content of gas in the serum of each group(P>0.05).Conclusion:Radish seed can increase Ghrelin and SP in serum,reduce NO and enhance intestinal propulsion,so as to promote gastrointestinal motility of rats with gastrointestinal heat retention,but Its effect is related to the radish seed dose.The promoting effect of large dose radish seed is more obvious.
