摘 要:目的探讨运动训练联合健心汤对心力衰竭大鼠GATA-4基因及蛋白表达的影响。方法清洁级SD大鼠140只分成:空白对照组、模型组、运动组、依普利酮组(10 mg/kg)、健心汤组(10 g/kg)、依普利酮(10 mg/kg)+运动组、健心汤(10 g/kg)+运动组。模型组、运动组、依普利酮组、健心汤组、依普利酮+运动组、健心汤+运动组建立心力衰竭模型。各组大鼠建模成功后给予相应药物及运动干预,空白对照组及模型组给予等体积的生理盐水。干预结束后,彩色多普勒超声诊断仪测定心功能指标,RT-PCR法及Western blotting法测定心肌GATA-4基因及蛋白水平。结果与空白对照组比较,模型组大鼠LVIDd、LVIDs水平、GATA-4 mRNA和蛋白水平升高,FS、EF水平降低(P <0.05);与模型组比较,运动组、依普利酮组、健心汤组、依普利酮+运动组、健心汤+运动组LVIDd、LVIDs水平、GATA-4 mRNA和蛋白水平降低,FS、EF水平升高(P <0.05);与运动组比较,依普利酮组、健心汤组LVIDd、LVIDs水平、GATA-4 mRNA和蛋白水平降低,FS、EF水平升高(P <0.05),且依普利酮组、健心汤组LVIDd、LVIDs、FS、EF、GATA-4 mRNA和蛋白水平相近(P> 0.05);依普利酮+运动组LVIDd、LVIDs水平、GATA-4 mRNA和蛋白水平低于依普利酮组、运动组,FS、EF水平高于依普利酮组、运动组(P <0.05);健心汤+运动组LVIDd、LVIDs、GATA-4 mRNA和蛋白低于健心汤组、运动组,FS、EF高于健心汤组、运动组(P <0.05);依普利酮+运动组和健心汤+运动组LVIDd、LVIDs、FS、EF、GATA-4 mRNA和蛋白水平相近(P> 0.05)。结论运动训练联合健心汤能改善大鼠心力衰竭程度,其效果优于单独运动训练、健心汤作用,其机制可能与运动训练联合健心汤抑制心力衰竭大鼠心脏GATA-4基因及蛋白表达水平有关。Objective:To investigate the effect of exercise training combined with Jianxin Decoction on expression of GATA-4 gene and protein in rats with heart failure. Methods:140 SD rats were divided into the control group,the model group,the exercise group,Eplidone group(10 mg/kg),Jianxin Decoction group(10 g/kg),Eplidone(10 mg/kg)+exercise group,Jianxin Decoction(10 g/kg)+exercise group. Except the control group,the other groups were established into heart failure models. After successful modeling,each group was given corresponding drugs and exercise intervention,while the control group and the model group were given equal volume of normal saline.At the end of the experiment,the cardiac function indicators were measured with color Doppler ultrasound,and the gene and protein levels of GATA-4 were measured with RT-PCR and Western blotting. Results:Compared with the control group,the levels of LVIDd,LVIDs,GATA-4 mRNA and protein in the model group increased,while the levels of FS and EF decreased(P < 0.05). Compared with the model group,the levels of LVIDd,LVIDs,GATA-4 mRNA and protein in other groups decreased,while the levels of FS and EF increased(P < 0.05). Compared with the exercise group,the levels of LVIDd,LVIDs,GATA-4 mRNA and protein in eplidone group and Jianxin Decoction group decreased,and those in eplidone group and Jianxin Decoction group were similar(P > 0.05),while the levels of FS and EF increased(P < 0.05). The levels of LVIDd,LVIDs,GATA-4 mRNA and protein in Eprinone+exercise group were lower,and the levels of FS and EF were higher than those in Eprinone group and exercise group(P < 0.05). The levels of LVIDd,LVIDs,GATA-4 mRNA and protein in Jianxin Decoction+exercise group were lower,and FS and EF were higher than those in Jianxin Decoction and exercise group(P < 0.05). The levels of LVIDd,LVIDs,FS,EF,GATA-4 mRNA and protein in the two groups were similar(P > 0.05).
