摘 要:在Chiralcel OD-H色谱柱上,采用正己烷与异丙醇或乙醇为流动相,对AEE788对映体进行了拆分,考察了流动相组成、柱温和流速等条件对拆分效果的影响。结果表明,AEE788对映体在正己烷-异丙醇-二乙胺(75:25:0.15,V/V/V)为流动相、柱温25℃、流速1.0 mL/min条件下,分离度为1.85,分析时间小于12 min。该分析方法的建立对药物AEE788不对称反应研究、分析/分离方法的开发及其生产过程的质量监控都具有非常重要的意义。Chromatographic model was employed for enantioseparation of AEE788 on the Chiralcel OD-H chiral column with hexane/alcohol(isopropyl alcohol or ethanol)mobile phase.The effects of chromatography conditions,such as mobile phase,temperature and flow rate to chiral recognition,had been investigated.The results indicated that a good separation was obtained at 25℃by using hexane-isopropyl alcohol-ethylenediamine(75:25:0.15,V/V/V)as the mobile phase with a flow rate of 1.0 mL/min.The analysis time was less than 12 min,and the resolution was 1.85.This method of HPLC had important significance in the real-time control of chiral intermediate in asymmetric reactions and production for AEE788.
