摘  要:目的观察中药开郁清热方对胰岛功能的作用以及其作用机制。方法选择INS-1细胞,通过高糖造成细胞功能衰竭模型,分别给予开郁清热方含药血清大、小剂量,并给予罗格列酮作为对照,观察各组大鼠在治疗前后胰岛素分泌、INS/IGF-1信号通路相关指标等的变化情况。结果开郁清热方对胰岛功能有一定的改善作用,在低糖环境下,胰岛分泌并无明显差异(P>0.05);但在高糖环境下,胰岛素水平明显增加(P<0.05)。开郁清热方能够提高胰岛细胞的活性,并具有升高IGF-1、Akt、PI3K。结论开郁清热方可以通过影响INS/IGF-1信号通路起到胰岛功能保护的作用。Objective To research the effects and its mechanism of Chinese herbal formula Kaiyu Qingre(KYQR)on islet function.Methods INS-1 cells with-cell function failure were induced by high glucose.At the same time,medicated serum of KYQRF with different doses was administered to the cells.Rosiglitazone was taken as a control to observe the changes in the level of insulin and the relevant INS/IGF-1 signal pathway in each group.Results Our data showed that KYQR had some effects on the insulin secretion.In a low glucose environment,no effective change in insulin secretion were observed(P>0.05);however,insulin levels increased significantly when INS-1 cells were exposed to a high glucose environment(P<0.05).KYQR could also enhance cell viability(P<0.05)in an effect similar to rosiglitazone.At the same time,KYQR could also increase the expression of IGF-1,Akt,PI3 K(P<0.05).Conclusion Our data suggests that KYQR can improve islet function by regulating the INS/IGF-1 signal pathway.
