摘 要:目的:探讨iPSc移植后对急性心肌梗死(AMI)小鼠心肌局部单相动作电位产生的影响,进而对iPSc移植的临床应用在电活动方面安全性进一步研究。方法:建立由美国癌症研究协会(institute of Cancer Research)命名的小鼠心肌梗死模型并将其随机分为四组。分别于移植iPSc各时间点检测各组小鼠心肌组织局部单向动作电位的变化。免疫组化染色法检测各组小鼠心肌缝隙连接蛋白43(Cx-43)的表达。结果:2周后,AMI+iPSc组小鼠心肌局部单相动作电位基本指标均较模型对照组明显回升,免疫组化结果显示AMI+iPSc组Cox-43的表达明显高于AMI组(P<0.05)。结论:iPSc移植后2周可明显改善梗死后小鼠的心肌局部单向动作电位基本指标,增强其电位稳定性。Objective:To investigate the effects of induced ploripoteat stem cells(iPSc)transplantation on the local myocardial monophasic action potential of aulte myocardial infarction(AMI)in mice,the clinical application of iPSc transplantation is further studied in the field of electrical activity.Methods:When AMI model mice named by American Institute of Cancer Research(ICR)was constructed successfully,They were divided into 4 groups randomly.The changes of local myocardial monophasic action potential in different groups of mice were checked at each time point after iPSc transplantation on cardiac tissue.The expression of connexin 43(Cx-43)in myocardium was checked by immunohistochemistry method in each group.Results:After two weeks,the basic indexes of local myocardial monophasic action potential in AMI+iPSc group mice were compared with the model control group was significantly increased.Immunohistochemistry result showed that the expression of Cox-43 in AMI+iPSc group was significantly higher than that in AMI group(P<0.05).Conclusions:After the iPSc transplantation two weeks,the basic indexes of local myocardial monophasic action potential was significantly improved after AMI and its potential stability was enhanced.
