摘 要:目的通过高脂血症大鼠模型制备方法的探索性研究,为正式建立大鼠高脂血症模型、开展山楂茶降脂效果研究奠定基础。方法选取SD雌性大鼠8只,平均体重(200±20)g,随机分为实验组与对照组,每组4只。实验组:自制50.7%高脂乳剂1.3 mL/(100 g·d),连续灌喂35 d;对照组:0.9%氯化钠溶液1.3 mL/(100 g·d),连续灌喂35 d。观察指标为体重、进食量、死亡情况,处死后观察肝脏质地、颜色、重量,进行血液生化检测、病理组织学观察。结果实验期间两组均无死亡;实验组与对照组体重比较差异无统计学意义(F=2.221,P=0.052);实验组进食量低于对照组,差异有统计学意义(F=162.012,P<0.001);实验组与对照组肝重比较差异无统计学意义(t=-0.906,P=0.400);血液检测:实验组与对照组的高密度脂蛋白(HDL-C)、血清中总胆固醇(TC)、低密度脂蛋白(LDL-C)含量差异有统计学意义(P<0.05),实验组与对照组TG的含量比较差异无统计学意义(P>0.05)。病理学检查:对照组肝细胞结构完整,大小均匀一致,细胞核形态规则、排列整齐,视野中无明显脂滴出现,而实验组35 d后肝细胞出现重度脂变,排列紊乱,形态大小各异,内充满大小不等的脂滴空泡,将细胞核挤向一侧,脂滴空泡主要集中于肝小叶周边部。结论本研究所采用方法能成功制备高脂血症大鼠模型。【Objective】To study the preparation method of hyperlipidemia rat model,and to establish the basis of the research on the effect of hawthorn substitute tea for reducing blood lipid.【Methods】Eight female SD rats were randomly divided into experimental group and control group,4 rabbits in each group,weight 200±20 g.The experimental group was fed with homemade 50.7% high-fat emulsion 1.3 ml/(100 g·d),for 35 days consecutively;the control group was fed with 0.9% sodium chloride solution1.3 ml/(100 g·d),for 35 days consecutively.The observation indexes were weight,food intake and death.The liver texture,color and weight were observed after the death,and blood biochemical test and histopathological observation were conducted.【Results】There was no death in the two groups during the experiment.There was no significant difference in weight between the experimental group and the control group(F=2.221,P=0.052).The food-intake of the experimental group was lower than that of the control group,and the difference was statistically significant(F=162.012,P<0.001).There was no significant difference in liver weight between the experimental group and the control group(t=-0.906,P=0.400).Blood test results showed that the contents of HDL-C,TC and LDL-C in the experimental group and the control group were statistically different(P<0.05),while the contents of TG in the experimental group and the control group were not statistically different(P>0.05).The results of pathology examination showed that in the control group,the structure of liver cells were integrity with uniform size,the cell nucleus was morphologically regular and neatly arranged,there was no obvious lipid droplets in the field of vision;in the experimental group,heavy grease appeared,liver cells were arranged at random with varied shapes and sizes and filled with lipid drops cavity,the nucleus was crushed to one side,the lipid drops around cavitation focuses on hepatic lobule.【Conclusion】The results of this study showed that the rat model of hyperlipide
