摘  要:目的探讨调控成年大鼠心肌细胞再生与调节性T细胞的关系。方法建立成年大鼠心肌再生模型,按照随机数字表法将大鼠分为研究组(行心尖切除术)和对照组(行假手术:只开胸不切除心尖组织),比较两组术后14 dFoxp3、CD25^+、CD4^+数,利用白喉毒素敲除T reg细胞,比较两组抑炎因子、转化生长因子、促炎因子的数量变化。结果研究组的Foxp3和CD25^+、CD4^+数明显多于对照组(t=3.643、8.510、11.992,P<0.05);研究组的抑炎因子(IL-10、IL-13)和转化生长因子(TGF-α)明显降低,促炎因子(IL-1β、IL-6)明显上升(t=5.810、3.692、18.999、11.110、12.397,P<0.05)。结论调节性T细胞对成年大鼠的心肌细胞有调控作用,能促进心肌细胞增殖,促进心脏修复,当调节性T细胞调节功能减弱时,大鼠体内的抑炎因子、转化生长因子将明显降低,促炎因子将明显上升,炎症反应加剧,最终导致大鼠的心肌细胞再生能力缺失。Objective To investigate the relationship between the regulation of adult rat cardiomyocyte regeneration and regulatory T cells.Methods An adult rat myocardial regeneration model was established.Rats were divided into a study group(with apical resection)and a control group(with sham operation:only open the chest without removing the apical tissue)according to the random number table method.The two groups were compared after surgery.14 dFoxp3,CD25^+,and CD4^+numbers were used to knock out T reg cells using diphtheria toxin and compare the changes in the numbers of anti-inflammatory factors,transforming growth factors,and pro-inflammatory factors between the two groups.Results The numbers of Foxp3,CD25^+and CD4^+in the study group were significantly higher than those in the control group.(t=3.663,8.510,11.992,P<0.05).The anti-inflammatory factors(IL-10,IL-13)of the study group.And transforming growth factor(TGF-α)were significantly reduced,and pro-inflammatory factors(IL-1β,IL-6)were significantly increased.(t=5.810,3.692,18.999,11.110,12.397 P<0.05).Conclusion Regulatory T cells can regulate the myocardial cells of adult rats,can promote the proliferation of cardiomyocytes,and promote cardiac repair.When the regulatory function of regulatory T cells is weakened,the anti-inflammatory factors and transforming growth factors in rats will be obviously decreased,the pro-inflammatory factors will rise significantly,and the inflammatory response will intensify,eventually leading to the loss of myocardial cell regeneration ability in rats.
