摘  要:目的:观察自身免疫性甲状腺炎(AIT)NOD.H-2h4小鼠microRNA-146(miR-146)的表达水平。方法:选取4周龄NOD.H-2h4雌性小鼠随机分为正常饮水(无菌双蒸水)组和高碘饮水(0.05%碘化钠水)组。甲状腺HE染色观察淋巴细胞浸润情况;获取CD4^+ T细胞及甲状腺组织small RNAs,RT-PCR检测miR-146表达水平;检测心、肝和肾中miR-146的表达水平。结果:高碘饮水组NOD.H-2h4小鼠甲状腺组织有不同程度的淋巴细胞浸润,甲状腺炎的发生率均高于对照组。与对照组相比,高碘饮水组小鼠脾脏CD4^+ T细胞和甲状腺中miR-146水平显著升高。高碘饮水组小鼠与对照组相比,心、肝和肾中miR-146的水平没有明显差别。结论:在NOD.H-2h4小鼠AIT模型中,甲状腺和脾脏CD4^+ T miR-146水平升高,可能参与自身免疫性甲状腺炎的发生和发展。Objective:To observe levels of miR-146 in NOD.H-2 h4 mice with iodine-induced autoimmune thyroiditis.Methods:Female NOD.H-2 h4 mice at 4 weeks were randomly divided into high-iodine group(0.05% NaI) and control group(sterile double distilled water).Thyroid sections were stained with HE assay.CD4^+T cells were collected by MACS.Small RNAs of CD4^+T cells and thyroid tissues were prepared for evaluating the levels of miR-146 by RT-PCR.The levels of miR-146 in heart,liver and kidney were also analyzed.Results:The present study found that the levels of miRNA-146 in CD4^+T cells and thyroid tissues of high-iodine group increased significantly than control group,while no difference of the levels of miRNA-146 in heart,liver and kidney was detected.Conclusion:MiR-146 may involve in the development of autoimmune thyroiditis in NOD.H-2 h4 mice.
