摘  要:目的建立大鼠脊髓损伤模型,观察大鼠在脊髓损伤后的运动功能变化,为临床前脊髓损伤的机制研究提供实验动物模型参考。方法 SD大鼠20只,随机分为对照组与手术给药组。手术暴露大鼠L2-3脊髓后,向髓内注射0.5%布比卡因,评价大鼠行为学改变及脊髓形态学改变。结果与对照组相比,术后1 d、7 d、14 d其BBB评分值分别为(15.33±1.02)分、(16.80±1.44)分、(17.28±1.07)分,低于相应同期对照组的BBB评分值,差异有统计学意义(t=5.436、3.620、4.756,P<0.05)。病理HE染色可见脊髓神经元出现空泡变性及坏死。结论大鼠脊髓腰麻针穿刺后注射布比卡因的大鼠动物模型,脊髓损伤效果具有可控的稳定性,利于实验室的再现性与重复性。Objective To establish a spinal cord injury model in rats and observe changes in motor function after spinal cord injury in rats, in order to provide a reference for experimental animal models for the study of the mechanism of preclinical spinal cord injury. Methods 20 SD rats were randomLy divided into control group and surgical administration group. After surgically exposing the L2-3 spinal cord to rats, 0.5% bupivacaine was injected intramedullarily to evaluate behavioral changes and spinal morphological changes in rats. Results Compared with the control group, the BBB scores were(15.33±1.02)points,(16.80 ± 1.44,)points and(17.28±1.07)pointsat 1 d, 7 d, and14 d after surgery, which were lower than the corresponding BBB scores of the corresponding control group,The results showed a significant difference(t=5.436,3.620,4.756,P<0.05);pathological HE staining showed vacuolar degeneration and necrosis of spinal neurons. Conclusion The rat animal model of bupivacaine injected with spinal cord anesthesia after spinal cord puncture in rats has a controllable and stable spinal cord injury effect, which is conducive to laboratory reproducibility and repeatability.
