摘 要:为研究西藏尼玛县羊传染性脓疱病毒遗传进化现状,采集了西藏尼玛县内某养殖户中疑似患羊传染性脓疱的患羊唇部分泌物,采用病毒分离、PCR、基因克隆、测序等技术,将分离株的B2L、F1L和ORF020基因与GenBank公布的部分流行株相应序列进行对比分析。结果显示,B2L基因在核苷酸水平显示96.4%~98%的同源性,与吉林分离株(MG712417)的同源性最高,为98%,与广东分离株(KY053526)的亲缘关系最近,同源性为97.9%;F1L基因在核苷酸水平显示95.3%~97.5%的同源性,与陕西分离株(KJ139957)的亲缘关系最近,同源性为97.5%;ORF020基因在核苷酸水平显示95.1%~99.5%的同源性,与陕西分离株(KJ139958)亲缘关系最近,同源性为99.5%。基于B2L、F1L、ORF020遗传进化分析表明,CPDV在不同地理环境和宿主中存在基因差异,但总体表现出保守性,同时表明西藏尼玛县境内CPDV流行的复杂性,研究结果可为该地区CPDV防控等相关研究提供参考。The purpose of this experiment was to research the genetic evolution of contagious pustular dermatitis virus,samples were collected from affected goats from raising households in Nyima County of Tibet,and virus isolation,PCR,cloning and sequencing technology were used.The results showed that the B2 Lgene shared 96.4%-98% nucleotide sequence homology,the highest homology with Jilin strain was98%,and the closest relationship with Guangdong strain was 97.9%;the F1 Lgene shared 95.3%-97.5%,the closest relationship with Shaanxi strain was 97.5%;the ORF020 gene shared 95.1%-99.5%,the closest relationship with Shaanxi strain was 99.5%.This further analyses of B2 L,F1 Land ORF020 genes proved that CPDV was conservative in different environments and hosts,also proved the complexity of CPDV in Nyima,Tibet,and provided theoretical basis for the related research of CPDV prevention and control in this area.
