胡桃楸青果皮指纹图谱及绿原酸含量测定研究

摘  要:目的建立胡桃楸青果皮的指纹图谱并测定其绿原酸的含量,结合化学计量学方法,对不同产地胡桃楸青果皮进行质量评价。方法采用Agilent Poroshell 120 SB-C18(2.1 mm×100 mm,2.7μm)色谱柱,以0.1%甲酸水(A)-乙腈(B)为流动相进行梯度洗脱,流速0.3 mL·min^-1,检测波长260 nm(指纹图谱检测)、325 nm(绿原酸测定),柱温30℃,建立23批不同产地胡桃楸青果皮的指纹图谱,并测定胡桃楸青果皮中绿原酸的含量;采用UHPLC-Q-TOF-MS/MS法,鉴定胡桃楸青果皮指纹图谱中的共有成分。结果绿原酸在0.350~166.5μg·mL^-1与峰面积线性关系良好(r=1.000),平均加样回收率为102.6%(RSD=2.3%);23批胡桃楸青果皮样品中确定了14个共有成分,并鉴定出其中7个成分;其相似度范围是0.795~0.989,聚类分析及主成分分析按产地将样品聚为3类,PLS-DA分析发现6个共有成分为区分不同产地样品的差异性成分。结论所建立的胡桃楸青果皮指纹图谱分析方法操作简单、专属性强,可为胡桃楸青果皮质量评价方法提供参考。Objective To establish the fingerprint of green walnut husks of Juglans mandshurica Maxim.,to determine the content of chlorogenic acid,and to evaluate the quality of green walnut husks of Juglans mandshurica Maxim.from different producing areas by chemometrics method.Methods Agilent Poroshell 120 SB-C18(2.1 mm×100 mm,2.7μm)chromatographic column was used to conduct gradient elution with 0.1%formic acid water(A)-acetonitrile(B)as the mobile phase at a flow rate of 0.3 mL·min^-1.The detected wavelength was 260 nm and 325 nm,and the column temperature was 30℃.Fingerprints of 23 batches of green walnut husks of Juglans mandshurica Maxim.from different origins were established.The content of chlorogenic acid in Juglans mandshurica was determined.UHPLC-Q-TOF-MS/MS method was used to identify the common components in the fingerprint of green walnut husks of Juglans mandshurica Maxim..Results The linearity of chlorogenic acid at 0.350-166.5μg·mL^-1 was good(r=1.000),and the average recovery rate was 102.6%(RSD=2.3%).Totally 14 common components were identified in 23 batches of green walnut husks of Juglans mandshurica Maxim.samples and 7 of them were identified.The similarity range was 0.795-0.989.Cluster analysis and principal component analysis grouped the samples into 3 categories.PLSDA analysis showed that 6 common components successfully distinguished samples from different origins.Conclusion The established fingerprint method of green walnut husks of Juglans mandshurica Maxim.is easy to operate and have strong specificity,which can provide a reference for the quality evaluation of green walnut husks of Juglans mandshurica Maxim..
