摘  要:为探索不同产地白术的遗传多样性及白术根茎、株高、叶形等关键农艺性状与ITS2条形码的关系,本研究收集了7个产区的白术根茎、种子和选种植株,共66份样品,通过提取白术药材DNA,PCR扩增ITS2序列并测序。并将ITS2序列与根茎形态、株高、叶形等指标进行相关性研究。结果表明白术ITS2序列比对后长度为229 bp,种内K2P遗传距离为0~0.013,共获得5种单倍型,其中A1和A2单倍型的频率分别为78.79%和13.64%,是频率最高的两种,分布范围最广,代表了绝大部分产地白术ITS2这个片段的遗传特征。单倍型与根茎性状或是栽培类型没有一一对应关系,但不同栽培类型,各单倍型出现的频率不同,狭叶类型比阔叶类型白术群体有更加丰富的变异。本研究发现,通过ITS2条形码技术与白术优株选育技术相结合,可以提高选种育种效率。In order to explore the genetic diversity of Atractylodes macrocephala from different origins and the relationship between ITS2 barcode and its key agronomic traits such as rhizome,plant height and leaf type,66 samples including rhizome,seed and selected plants of Atractylodes macrocephala from 7 producing origins were collected.After DNA extracting,ITS2 sequence was amplified by PCR and sequenced.Finally,the relationship between ITS2 sequence and rhizome morphology,plant height and leaf shape was studied.The results indicated that five haplotypes were obtained after ITS2 sequence alignment of Atractylodes macrocephala with a length of 229 bp and the intraspecific K2P genetic distance was 0~0.013.A1 and A2 haplotype,the most frequent and widely distributed haplotypes,whose frequencies were 78.79%and 13.64%,respectively,represented the genetic characteristics of ITS2 fragment in Atractylodes macrocephala.Although there was no one-to-one correspondence between haplotypes and rhizome traits or cultivation types,the occurrence probability of haplotypes was distinct in different cultivation types.The narrow-leaved type had more abundant variation types than the broad-leaved Atractylodes macrocephala population.All in all,the combination of ITS2 barcode technology and the breeding techno-logy of Atractylodes macrocephala can improve the efficiency of breeding.
