摘 要:目的研究酒炙前后当归多糖对血瘀证大鼠的作用,探讨其可能的机制。方法将SD大鼠随机分为空白组,模型组,复方丹参滴丸组(72.9 mg·kg^-1·d^-1),生当归多糖低、高剂量组(400、800 mg·kg^-1·d^-1),酒当归多糖低、高剂量组(400、800 mg·kg^-1·d^-1),连续灌胃15 d后,采用皮下注射盐酸肾上腺素和冰水浴共同复制急性血瘀大鼠模型,通过自动血流变检测仪测定全血黏度(WBV)和血浆黏度(PV),检测大鼠血液流变学指标的变化;通过全自动血凝仪测定血浆的凝血四项,即凝血酶原时间(PT)、活化部分凝血酶原时间(APTT)、凝血酶时间(TT)、纤维蛋白原(FIB),检测大鼠凝血功能的变化;HE染色法观察大鼠心、肺组织病理情况;测定血浆中超氧化物歧化酶(SOD)、丙二醛(MDA)和谷胱甘肽过氧化物酶(GSH-Px)的含量,检测大鼠氧化应激水平的变化。结果与模型组比较,生当归多糖高剂量组和酒当归多糖低、高剂量组均能降低WBV和PV水平,降低血浆中FIB和MDA含量,延长血浆的PT、APTT和TT值,增强血浆中SOD和GSH-Px的活性(P <0.05,P <0.01),改善心肌细胞加宽、心肌细胞核变圆、肺泡壁增厚、肺间质水肿和肺出血等现象。与复方丹参滴丸组比较,生当归多糖组改善大鼠血液流变性和凝血系统功能的效果均与之接近,差异无统计学意义(P> 0.05),而酒当归多糖高剂量组的效果均优于复方丹参滴丸组(P <0.05)。结论生当归多糖及酒当归多糖均可改善急性血瘀大鼠的血液流变性、凝血系统功能和病理损伤程度,其中酒当归多糖的作用优于生当归多糖,其机制可能与改善大鼠氧化应激水平有关。Objective To study the effect of Angelica polysaccharides on blood stasis rats before and after wine broiled,and to explore its possible mechanism. Methods SD rats were randomly divided into blank group,model group, compound Danshen dripping pill group(72.9 mg · kg^-1· d^-1), raw Angelica polysaccharide groups(400,800 mg·kg^-1·d^-1),and wine broiled Angelica polysaccharide groups(400,800 mg·kg^-1·d^-1). After 15 days of continuous intragastrically administered drugs,the acute blood stasis rat model was co-produced by subcutaneous injection of adrenaline hydrochloride and ice water bathing, and the whole blood viscosity(WBV) and plasma viscosity(PV)were measured by an automatic hemorhematometer to detect the variety of hemorheology indicators in rats;automated blood coagulation analyzer was engaged to measure the four indexes of plasma coagulation,namely prothrombin time(PT),activated partial prothrombin time(APTT),thrombin time(TT),fibrinogen(FIB),to detect coagulation function of rats. HE staining was used to visualize the pathological changes of heart and lung tissues in rats;the activities of superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px), and the content of malondialdehyde(MDA)in plasma were determined to represent oxidative stress level in rats. Results Compared with the model group,the high dose group of raw Angelica polysaccharide and wine broiled Angelica polysaccharide groups can reduce the whole blood viscosity(WBV)and plasma viscosity(PV),reduce the content of FIB and MDA in the plasma,prolong the PT,APTT and TT values in the plasma,enhance the activities of SOD and GSH-Px in the plasma(P < 0.05,P < 0.01),improve the symptoms of myocardial cell widening,cardiac nucleus rounding,alveolar wall thickening, pulmonary interstitial edema and pulmonary hemorrhage. Compared with the compound Danshen dropping pill group,the effect of raw Angelica polysaccharide groups on improving the hemorheology and coagulation system of rats is close to it,the difference was not statistically significant(
