摘  要:目的观察香舒丸对乳腺增生大鼠的改善作用,探讨其治疗作用的性激素调节机制。方法将雌性SD大鼠随机分为空白对照组、模型组、他莫昔芬组(3 mg·kg^-1)及香舒丸低、中、高(2.26、4.52、9.04 g·kg^-1)剂量组,每组10只。肌肉注射苯甲酸雌二醇与黄体酮复制乳腺增生大鼠模型。大鼠灌胃给药,每天1次,连续90 d。以游标卡尺测量大鼠第2对乳头的高度、直径;HE染色观察乳腺组织病理形态学变化;ELISA法检测血清中雌二醇(E2)、孕酮(P)、泌乳素(PRL)含量;免疫组化法及Western Blot法检测乳腺组织中雌激素受体(ERα+β)、孕激素受体(PR)表达情况。结果与空白对照组比较,模型组大鼠乳房直径和乳头高度明显增加(P <0.01);乳腺小叶体积增大,小叶内腺泡数量增多,导管扩张,腺上皮增厚;血清中E2、PRL含量明显升高(P <0.01),P含量明显降低(P <0.01);乳腺组织中ERα、PR蛋白表达明显上调(P <0.01),ERβ蛋白表达明显下调(P <0.01)。与模型组比较,香舒丸各剂量组大鼠乳头高度和直径均明显下降(P <0.01);乳腺小叶腺泡数减少,导管直径明显减小;血清中E2与PRL含量明显降低(P <0.01),P含量明显升高(P <0.01);乳腺组织中ERα、PR蛋白表达明显下调(P <0.05,P <0.01),ERβ蛋白表达明显上调(P <0.01)。结论香舒丸可能通过调节性激素水平和乳腺组织中雌、孕激素受体的表达,从而发挥抗大鼠乳腺增生的作用。Objective To investigate the effect of Xiangshu pill on sex hormone levels in rats with hyperplasia of mammary gland. Methods SD rats were randomly divided into 6 groups,including normal control group,model group,positive control group(3 mg·kg^-1 Tamoxifen),Xiangshu pill groups(2.26,4.52,9.04 g·kg^-1). Rats were consecutively injected with estradiol benzoate and progestin to establish the hyperplasia of mammary gland(HMG)model. Then the drugs were orally administered for 90 days. After administration,nipple’s height and diameter were measured. The histopathological changes of mammary gland tissue were detected by HE staining. Serum sex hormone levels were measured by ELISA. Immunohistochemistry staining and Western Blot were performed to evaluate the expression levels of estrogen receptor(ERα+β)and progesterone receptor(PR). Results Compared with the normal control group, nipple’s height and diameter in model group were increased significantly(P < 0.01). Mammary epithelial tissue had significantly proliferative lesions,including hyperplasia in most lobules,increased numbers of acini and ducts,and thickened glandular epithelium. E2 and PRL contents were obviously increased in serum,as well as ERα and PR in mammary gland(P < 0.01). P content and ERβ expression were notably decreased(P <0.01). However, Xiangshu pill significantly reduced nipple’s height and diameter(P < 0.01). Xiangshu pill improved the pathological features of mammary gland. Xiangshu pill not only markedly attenuated the over-expression of E2 and PRL in serum as well as ERα and PR in mammary gland(P < 0.05,P < 0.01),but also up-regulated P level(P < 0.01)in serum and ERβ expression in mammary gland(P < 0.01). Conclusion Xiangshu pill might exert anti-hyperplasia effects in rats via modulating serum sex hormone levels and mammary expression of related receptors.
