摘  要:目的观察五指毛桃对吲哚美辛所致大鼠胃黏膜损伤的修复作用,并探讨其作用机制。方法皮下注射吲哚美辛复制大鼠胃黏膜损伤模型。将42只SD雄性大鼠随机分为正常组,模型组,五指毛桃水提物高、低剂量组(14、7 g·kg^-1)。灌胃给药后,肉眼观察胃黏膜损伤情况;胃黏膜组织HE染色后显微镜下观察其病理改变,并计算其损伤病理评分;免疫组化法检测胃黏膜紧密连接蛋白(Claudin-1、Occludin、ZO-1)、黏附连接蛋白(E-cadherin、α-catenin、β-catenin)、胃肠道特异性转录因子(SOX2、CDX2)表达。结果五指毛桃高剂量组大鼠胃黏膜损伤大体评分和病理评分均低于模型组(P <0.05);五指毛桃高剂量组能提高模型大鼠胃黏膜紧密连接蛋白(Claudin-1、Occludin、ZO-1)、黏附连接蛋白(E-cadherin、α-catenin)及转录因子SOX2表达,与模型组比较差异有统计学意义(P <0.05)。结论五指毛桃有防治吲哚美辛所致大鼠胃黏膜损伤的作用,作用机制与其影响胃黏膜紧密连接和黏附连接蛋白及转录因子SOX2的表达有关。Objective To evaluate the effects of Wuzhimaotao,a Lingnan traditional Chinese medicine,on gastric mucosal injury induced by indomethacin in rats, and to further explore the potential mechanisms. Methods Rat models with gastric mucosal injury were induced by subcutaneously injection with indomethacin. Forty two SD rats were randomized into control group,model group,Wuzhimaotao decoction high and low dose groups(14 and 7 g·kg^-1).After intragastricly administration with correspondent drugs,rats were sacrificed. Overall gastric mucosal injury in each group was scored according to naked eye observation. The pathological changes of gastric mucosa injury were observed and scored under microscope after HE staining. Immunohistochemisty were carried out to visualize the expression of tight junction proteins(Claudin-1,Occludin,ZO-1),adhesion junction proteins(E-cadherin,α-catenin, β-catenin) and related transcription factors(SOX2, CDX2) in gastric mucosa. Results Macroscopic scores and pathological scores of Wuzhimaotao group(14 g·kg^-1)were lower than those in model group(P < 0.05).Wuzhimaotao group(14 g · kg^-1) also improved the expression of tight junction proteins(Occludin, Claudin-1,ZO-1),adhesion junction proteins(E-cadherin,α-catenin)and transcription factor(SOX2)in gastric mucosa of model rats(P < 0.05). Conclusion Wuzhimaotao decoction can prevent and treat gastric mucosal injury induced by indomethacin,the mechanism is associated with the regulation of tight junction proteins,adhesion junction proteins and specific transcription factor expression.
