摘 要:目的研究复方鱼腥草对高糖高脂联合低剂量链脲佐菌素(STZ)诱导的2型糖尿病大鼠的治疗作用。方法利用高糖高脂饲料联合腹腔注射低剂量STZ建立2型糖尿病(T2DM)大鼠模型。然后,将模型复制成功大鼠随机分为罗格列酮组、复方鱼腥草组和模型组,另设正常对照组。各给药组大鼠灌胃给予相应的药物,正常组与模型组仅给予等量0.5%的羧甲基纤维素钠(CMC-Na)4 mL·kg^-1溶液,每周给药6 d,共8周。药物治疗过程中观察大鼠的体质量、饮水量以及饮食量并记录数据,用尾部采血法检测大鼠空腹血糖值(FBG)。治疗8周后,大鼠进行腹主动脉取血并且测定其各项血液生化指标。采用不同的试剂盒分别测定空腹胰岛素(FINS)、胰高血糖素样肽-1(GLP-1)、糖化血清蛋白(GSP)、还原型谷胱甘肽(GSH)。结果在给予相应药物治疗后,各治疗组糖尿病大鼠的空腹血糖、体质量、水和食物摄入量均有改善。复方鱼腥组能明显降低大鼠血清肌酐(Scr)(P <0.05)、尿素氮(BUN)(P <0.05)、血尿酸(SUA)(P <0.05)和GSP水平(P <0.01)。复方鱼腥组能明显提高FINS水平和GLP-1(P <0.05,P <0.05)以及降低胰岛素抵抗指数(HOMA-IR)(P <0.05)。结论复方鱼腥草对T2DM大鼠有较好的治疗作用,通过改善各项血液生化指标,降低FBG水平并改善血清GLP-1和FINS水平以及胰岛素抵抗发挥作用。Objective To investigate the therapeutic effect of Compound Houttuynia herba on type 2 diabetic mellites(T2DM)rats induced by high-sugar and high-fat diet plus low-dose streptozotocin(STZ). Methods At first,T2DM rat model was established by high-sugar and high-fat diet combined with low-dose STZ,T2DM model rats were randomly divided into 3 group: Rosiglitazone(Rsg) group, model group, Compound Houttuyniae herba(CH)group;a normal control(Normal) group was also set. Rats in each treatment group were given corresponding medicine by gavage administration, the Normal group and model group were given same dose of 0.5% sodium carboxymethyl cellulose(CMC-Na),6 days a week for 8 weeks. The body weight,water intake and dietary intake of rats were observed and recorded at 2, 4, 6 and 8 week, and fasting blood sugar(FBG) were detected by the portable blood glucose detection. At the eighth week, the blood was taken from the abdominal aorta for blood biochemical tests. Glucagon-like peptide-1(GLP-1),glycated serum protein(GSP)reduced glutathione(GSH)and fasting insulin(FINS)were tested using correspondent commercial kits. Results The FBG,body weight,water and food intake of diabetic rats in each drug treatment group were improved after giving corresponding medicine. CH group could significantly reduce the levels of serum creatinine(Scr)(P < 0.05),blood urea nitrogen(BUN)(P <0.05),uric acid(SUA)(P < 0.05)and GSP(P < 0.01)in serum of rats. And CH group have the effect of improving the level of FINS and GLP-1(P < 0.05, P < 0.05), and decreasing homeostasis model assessment of insulin resistance(HOMA-IR)(P < 0.05). Conclusion Compound Houttuynia herba(CH)have better treatment effect on T2DM rats,which can improve the blood biochemical indexes and insulin resistance,regulate FBG and elevate the levels of serum GLP-1 and FINS in T2DM rats.
