摘 要:目的探讨糖络宁对糖尿病周围神经病变(DPN)大鼠背根神经节(DRG)中Toll样受体2/4(Toll 2/4)表达的调节作用。方法选用雄性8周龄SD大鼠80只,随机选取14只大鼠为正常组,余66只大鼠腹腔单次注射链脲佐菌素60 mg/kg,诱导DPN大鼠模型,其中61只造模成功。采用随机数字表法分为模型组(21只)、西药组(19只)、中药组(21只)。中药组灌胃糖络宁5 g/(kg·d),西药组灌胃α-硫辛酸20 mg/(kg·d),正常组、模型组灌胃等量蒸馏水,连续给药8周。Western blot法检测DRG中TLR2、TLR4蛋白表达水平。结果正常组大鼠TLR2、TLR4蛋白表达水平较弱,模型组TLR2、TLR4表达水平高于正常组(P<0.01);中药组、西药组TLR2和TLR4蛋白表达水平低于模型组(P<0.05);中药组TLR2、TLR4蛋白表达水平与西药组比较,差异无统计学意义(P>0.05)。结论糖络宁可能通过抑制TLR2、TLR4蛋白表达,影响其介导的NF-κB等信号通路,降低炎症及氧化应激,减少神经损伤。Objective To investigate the regulating effect of Tangluoning on Toll-like receptors 2/4(TLR2/4)in dorsal root ganglion(DRG)of rats with diabetic peripheral neuropathy(DPN).Methods Eighty of eight-week-old male SD rats were selected.Fourteen rats were randomly selected as normal group,the rest of 66 rats were intraperitoneally injected with 60 mg/kg streptozotocin to induce DPN rat model,among which 61 rats were succeeded.They were divided into the model group(21 rats),the Western medicine group(19 rats)and the Chinese medicine group(21 rats)by random number table method.Chinese medicine group was gavaged Tangluoning with 5 g/(kg·d),the Western medicine group was gavagedα-lipoic acid with 20 mg/(kg·d),the model group and normal group were gavaged equal amount of distilled water.The drug was administered continuously for 8 weeks.After 8 weeks,the expression of TLR2 and TLR4 proteins in DRG was detected by Western blot.Results The expression levels of TLR2 and TLR4 proteins in normal group were weak,while the expression levels of TLR2 and TLR4 proteins in the model group were higher than those in normal group(P<0.01).The expression levels of TLR2 and TLR4 proteins in the Chinese medicine group and the Western medicine group were lower than those in the model group(P<0.05).There were no statistically significant differences in the expression levels of TLR2 and TLR4 proteins between the Chinese medicine group and the Western medicine group(P>0.05).Conclusion Tangluoning may have impact on the signaling pathways such as the mediated NF-κB by inhibiting the expression of TLR2 and TLR4,thereby reducing inflammation and oxidative stress and reducing nerve damage.
