摘 要:【目的】探讨香蕉皮多酚(banana peels polyphenols)对D-半乳糖(D-galactose,D-gal)致衰大鼠皮肤的影响.【方法】将SD大鼠随机分成对照组、衰老模型组和香蕉皮多酚组,采用D-gal构建衰老大鼠模型,观察大鼠的生长状况,HE染色法观察皮肤组织病理变化,ELISA法检测Ⅰ、Ⅱ型胶原蛋白含量,吖啶橙/溴化乙啶(AO/EB)荧光双染法检测细胞凋亡,实时荧光定量PCR和Western-blotting检测皮肤组织基质金属蛋白酶-1(matrix metalloproteinases-1,MMP-1)、水通道蛋白3(aquaporin3,AQP3)mRNA水平和蛋白含量.【结果】与衰老模型组比较,香蕉皮多酚干预后,大鼠皮肤病理改变明显减轻;细胞凋亡率显著降低,Ⅰ、Ⅱ型胶原蛋白含量显著升高,MMP-1基因表达水平及蛋白含量下降,AQP3基因表达水平及蛋白含量则增加.【结论】D-gal可致皮肤结构损伤和功能衰退,香蕉皮多酚能拮抗D-gal,从而减轻对皮肤的损伤,其机制可能与增加胶原蛋白含量、抑制皮肤细胞凋亡、调节MMP-1和AQP3基因的表达有关.【Objective】 To study the effect of banana peel polyphenols on the skin of D-galactose induced aging rats.【Method】 SD rats were divided into control group,aging model group and banana peel polyphenols treated group.Aging rat models were made by D-gal.The growth of the rats was monitored.The pathological changes of skin were analyzed by HE staining.ELISA was used to determine the contents of typeⅠand type Ⅱ collagen.The cell apoptosis were detected by AO/EB double stained method.Real-time PCR and Western-blotting were used to detect mRNA and protein content of MMP-1 and AQP3 in skin tissue.【Result】 Compared with aging model group,the intervention of banana peel polyphenols could obviously alleviate skin pathological changes.The levels of cell apoptosis were obviously reduced.The contents of typeⅠand type Ⅱ collagen were obviously increased.The expressions of MMP-1 gene were down-regulated,while expressions of AQP3 gene were statistically up-regulated.【Conclusion】 D-gal can induce structural and functional injury of the skin.Banana peel polyphenols can antagonize D-gal to reduce the damage to skin.The mechanism may be associated with enhancing collagen contents,inhibiting skin cell apoptosis,regulating expressions of gene MMP-1 and AQP3.
