摘  要:几丁质是N乙酰葡糖胺通过β1,4糖苷键连接聚合而成的结构多糖,在医药、食品、纺织、环保等领域具有重要应用价值。以金针菇菌渣为基料,开展绿色木霉的培养基配方优化及发酵试验,以提高真菌中几丁质含量。结果表明:绿色木霉菌的最适碳源、氮源分别为葡萄糖30 g/L和硫酸铵4 g/L,金针菇菌渣100 g/L,在初始pH为6.0、培养基装液量为50 mL(250 mL三角瓶)、发酵周期为3 d时,几丁质含量达到最高(13%),质量浓度可达到2.65 g/L。本结果为低成本、工业化生产几丁质提供了科学依据。Chitin is a structural polysaccharide formed by the polymerization of N-acetylglucosamine byβ-1,4-glycosidic linkage.It has shown great application potential in pharmacy,food,textile and environmental protection.We optimized the culture medium for Trichoderma viride with spent substrate of Flammulina velutipes as the basic raw material,to increase the chitin content.The optimum ingredientsare medium compostion was 30 g/L glucose,4 g/L ammonium sulphate,and 100 g/L spent substrate of Flammulina velutipes.We obtained the highest content of chitin of 13%(titer of 2.65 g/L)in the 3rd day of the fermentation,with initial pH of 6 and medium volume of 50 mL in a 250-mL flask.This study provides scientific support to commercially producing chitin in a low-cost way.
