摘  要:目的基于网络药理学探讨国医大师徐景藩抑肝扶脾方治疗肠易激综合征(IBS)的“多因微效”作用机制。方法利用网络药理学将抑肝扶脾方有效化合物成分及其所作用的基因、蛋白质,与IBS的生物分子学信息进行网络映射,以获取两者关联的靶基因、靶蛋白;在网络上对IBS与抑肝扶脾方的关系进行机制性计算以获取关键靶标;对关键靶标的功能及互作关系进行分析与预测,并以网络可视化图的形式呈现。结果IBS与抑肝扶脾方关联靶基因21个,靶蛋白202个,关键靶标11个。通路富集主要作用于海马信号、神经活性配体受体交互作用、含血清素的神经突触等神经信号相关通路;皮质醇合成与分泌,胰腺分泌、蛋白消化与吸收等内分泌信号相关通路;ABC转运蛋白、丝裂原活化蛋白激酶、核因子-κB、NOD样受体、过氧化物酶体增殖剂激活受体、Wnt信号、Th17细胞分化等免疫炎症相关信号通路。结论国医大师徐景藩抑肝扶脾方对IBS生物分子网络具有多水平、多层次的调控作用,该方可能通过影响IBS的功能基因网络,在干预本病有关生物分子及相关基因表达上,发挥“多因微效”的整体调节作用。Objective To study underlying mechanisms of Yigan Fupi Recipe(YGFPR)by Prof.XU Jing-fan,the National Master of traditional Chinese Medicine(TCM),in treating irritable bowel syndrome(IBS)based on network pharmacology.Methods Network pharmacology was used to map the active compounds in YGFPR and the genes and proteins it acts on to the biological molecular network of IBS.With the target genes and proteins obtained,the relationship between IBS and YGFPR was calculated by certain parameters on the network in order to acquire the key targets.The functions and interactions of these key targets were analyzed and predicted through pathway enrichment,and results were shown in the form of network visualization.Results The numbers of associated target genes,target proteins,and key targets with IBS from YGFPR were 21,202,and 11,respectively.Three main pathways associated with neural signals were enriched:hippo signal,neuroactive ligand-receptor interaction,and serotonergic synapse.Two main pathways associated with endocrine signals were enriched:cortisol synthesis and secretion,and protein digestion and absorption.Seven main pathways associated with immune signals were enriched:ATP-binding cassette(ATP)-binding cassette(ABC)transporters,mitogen activated protein kinase(MAPK)signal,nuclear factor-kappa B(NK-κB)signal,NOD-like receptor signal,peroxisomal proliferators activate receptors(PPAR)signal,Wnt signal and Th17 cell differentiation.Conclusions YGFPR by Prof.XU Jing-fan showed a regulatory function on biological molec-ular network of IBS at multilevel ways.It played a holistic role of mild effects of multi-factors in intervening biological molecules and gene expressions associated with IBS by affecting its functional genetic network.
