摘 要:为了挖掘大丽轮枝菌β-1,4-内切木聚糖酶基因,通过生物信息学手段对其进行基因组水平鉴定和系统进化及表达模式分析。结果表明:从大丽轮枝菌全基因组数据库鉴定到11个β-1,4-内切木聚糖酶基因,其中4个属于糖苷水解酶10家族,5个属于11家族,2个属于43家族。11个基因随机分布在5个染色体上,其中3号染色体上包含4个基因,形成1个基因簇;其他染色体上仅含有1~2个基因,未形成基因簇。转录组测序表明,VDAG_03790、VDAG_03808、VDAG_03809、VDAG_05043和VDAG_06165这5个基因在棉花感病品种根系分泌物培养的大丽轮枝菌中的表达量明显高于其在耐病品种根系分泌物和水培养的样本中的表达量,表明这些基因对棉花感病品种的根系分泌物有明显的响应,暗示它们可能与大丽轮枝菌的致病性相关。In order to explore the Endo-β-1,4-xylanase genes in Verticillium dahliae,bioinformatics method was used to identify the Endo-β-1,4-xylanase genes from the genomic level,and to analyzeits phylogenetic and expression patterns.The results showed that 11 Endo-β-1,4-xylanase genes were identified from the whole genome database of V.dahliae.Among them,4 Endo-β-1,4-xylanase genes belonged to 10 families of glycoside hydrolase,5 belonged to the 11 family and 2 belonged to the 43 family.Eleven genes were randomly distributed on five chromosomes,including 4 genes on chromosome 3,which formed a gene cluster,only 1-2 genes on other chromosomes,which did not form a gene cluster.The transcriptome sequencing results showed that the expression levels of VDAG_03790,VDAG_03808,VDAG_03809,VDAG_05043,and VDAG_06165 in V.dahliae cultured with root exudate from susceptible cultivars were significantly higher than that in root exudates from cultivars resistant to diseases and water samples,the results indicated that these genes had obvious response to the root exudates from susceptible strains,this might be related to the pathogenicity of V.dahliae.The experiment may provide a foundation for further study on role of Endo-β-1,4-xylanase gene in the pathogenicity and the molecular mechanism of pathogenicity of V.dahliae.
