摘 要:该研究利用荧光SSR标记构建了9个柳树新品种的指纹图谱。11对SSR标记共检测到56条等位片段,每个位点等位基因数3—8不等,平均为5.1个。观测杂合度(Ho)变化范围为0.3333—1.0000,平均为0.6875;期望杂合度(He)变化范围为0.4771—0.8824,平均为0.6263;多态信息量(PIC)变化范围为0.4212—0.8134,平均为0.6059。优选的3对核心引物中,SALeSSR0086与SALeSSR0733组合、SALeSSR0086与SALeSSR0920组合可完全区分9个柳树新品种。聚类结果显示,9个柳树新品种遗传相似系数在0.6059与0.9107之间。该研究建立的荧光SSR基因分型体系高效、准确,不仅能为柳树品种鉴定和新品种保护提供科学理论依据,也能为柳树进一步育种工作奠定坚实的基础。The fingerprint of 9 Salix varieties was established by fluorescent SSR marker in this study.56 alleles were detected by 11 pairs of SSR markers from Salix,and the number of alleles per locus ranged from 3 to 8,with an average of 5.1 alleles.The observed heterozygosity(Ho),the expected heterozygosity(He)and the polymorphic information content(PIC)varied from 0.3333—1.0000(mean 0.6875),0.4771—0.8824(mean 0.6263)and 0.4212—0.8134(mean 0.6059),respectively.Among the 3 optimal pairs of core primers,SALeSSR0086 and SALeSSR0733 combinations,SALeSSR0086 and SALeSSR0920 combinations could completely distinguish 9 Salix varieties.The cluster analysis showed that the similarity coefficient of the 9 Salix varieties ranged from 0.6059—0.9107,and the different varieties from the same parents could be clustered in the same branch.The fluorescence SSR genotyping system established in this study was efficient,rapid and accurate,which could not only provide scientific theoretical basis for the identification of varieties and the protection of new Salix varieties,but also provide a solid foundation for the further Salix breeding.
