摘 要:目的探索电针对佐剂性关节炎大鼠镇痛抗炎效应的影响。方法两批Wistar大鼠随机分为盐水组、模型组和电针组(n=15/n=12),于右后足足底注射完全弗式佐剂(CFA)以制备动物模型。电针双侧"足三里穴""环跳穴"治疗佐剂性关节炎大鼠,以大鼠足底热辐射痛阈(n=15)和膝关节IL-2、TNF-α(n=12)等炎性因子水平变化作为疗效观察指标,观察针刺不同时间点对大鼠镇痛抗炎效应的影响。结果电针后即刻和24 h后大鼠热辐射缩足时间均显著升高,与模型组相比,电针后即刻痛阈变化在1d、5 d、10 d、20 d有统计学差异(P<0.05),电针24 h后痛阈在1 d、5 d、10 d有统计学差异。CFA致炎后,IL-2、TNF-α释放增多。经电针治疗后IL-2、TNF-α释放减少,各时间点电针组与模型组比较无统计学差异。结论电针治疗对佐剂性关节炎大鼠有显著的镇痛作用,抗炎作用尚未发现显著效应,有待进一步研究。Objective To observe the effect of acupuncture on the analgesic and anti-inflammatory effect of adjuvant arthritis rats.Methods Wistar rats were randomly divided into the saline group,the model group and the electroacupuncture group(n=15/n=12).CFA was injected into the right hind foot to make arthritis model.We used electroacupuncture on both sides of"Zusanli(ST36)"and"Huantiao(GB30)"in the treatment of adjuvant arthritis rats.The changes of the paw withdrawal latency(PWL)(n=15)and the level of IL-2 and TNF-α(n=12)in the knee joint were observed to evaluate the effect of acupuncture on analgesic and anti-inflammatory effects at different time points in rats.Results The time of PWL was significantly increased immediately after electroacupuncture and 24 h after electroacupuncture.Compared with the model group,the immediatepain threshold changes were statistically different 1 d,5 d,10 d and 20 d after electroacupuncture(P<0.05),the pain threshold 24 h after electroacupuncturewas statistically different1 d,5 d and 10 d.After CFA inflammation,IL-2 and TNF-αrelease increased.The levels of IL-2 and TNF-αwere decreased after electroacupuncture treatment.But there was no significant difference between the electroacupuncture group and the model group at each time point.Conclusion Electroacupuncture treatment has significant analgesic effect on adjuvant arthritis rats,but the anti-inflammatory effect is not foundtobe significant,which needs further study.
