摘  要:目的检测RhoA在肝细胞胰岛素抵抗状态下表达量的变化,并对RhoA与胰岛素抵抗的相关性进行分析。方法采用蛋白印迹技术检测RhoA在正常对照组和肝细胞胰岛素抵抗组的表达差异,采用生物信息学方法对RhoA与胰岛素信号通路相关蛋白的相互作用进行分析。结果肝细胞胰岛素抵抗组中RhoA表达显著高于正常对照组(P<0.05)。蛋白质相互作用分析表明RhoA直接与PIK3CA、ROCK1和PTEN相互作用。结论RhoA可能通过与PIK3CA、ROCK1和PTEN的相互作用,影响胰岛素信号通路PI3K/AKT的正常转导,引发胰岛素抵抗。Objective To detect expression of RhoA in liver cells with insulin resistance,and analyze the correlation between RhoA and insulin resistance.Methods The expression difference in RhoA between the normal control group and the hepatocyte insulin resistance group was detected by Western blot,and the interaction between RhoA and the protein on insulin signaling pathway was analyzed by bioinformatic methods.Results The expression of RhoA in the hepatocyte insulin resistance group was significantly higher than that in the normal control group(P<0.05).Protein-to-protein interaction analysis showed that RhoA directly interacts with PIK3CA,ROCK1 and PTEN.Conclusion RhoA may affect the normal transduction of PI3K/Akt signaling pathway through its interaction with PIK3CA,ROCK1 and PTEN,leading to insulin resistance.
