摘 要:大肠埃希氏菌(Escherichia coli,E.coli)又称大肠杆菌,因结构简单、遗传背景明确而成为基因克隆实验中最常用的受体菌种。感受态细胞的制备与转化是生物转化实验中的重要环节,大肠杆菌感受态细胞的转化效率直接影响携带目的基因的重组质粒能否导入受体细胞,表达出新的遗传性状。大肠杆菌感受态细胞(competent cells)制备常采用物理或化学方式改变细胞膜通透性,诱导受体菌摄取外源性DNA并整合入受体菌。此外,细菌细胞密度值(OD600)、转化液浓度、热激参数、培养条件等因素也影响转化效率。综述了国内外大肠杆菌感受态细胞的制备及转化方法,以期为各大实验室制备和转化大肠杆菌感受态细胞提供参考。Characteristics of simple structure and clear genetic background has made Escherichia coli(E.coli)becoming the most commonly used receptor strain in gene cloning.Preparation and transformation of competent cells is an important part of biotransformation experiments.Transformation efficiency of E.coli competent cells directly affects whether the recombinant plasmid carrying the target gene can be introduced into the recipient cell and express new genetic traits.Physical or chemical means are often used to change cell membrane permeability for preparation of E.coli competent cells,which induces recipient bacteria to take up exogenous DNA and integrates it into recipient bacteria.In addition,factors such as bacterial cell density(OD600),concentration of transformant,heat shock parameters,and culture condition also affect transformation efficiency.Preparation and transformation methods of E.coli competent cells at home and abroad were reviewed,to provide reference for preparation and transformation of E.coli competent cells by major laboratories.
