摘 要:目的:探究基于SHED聚合体的治疗大鼠脊髓损伤的有效疗法。方法:建立大鼠脊髓全横断损伤模型(n=28),分为4组:SHED聚合体组(SHED-CA,n=8),SHED聚合体联合SHED悬液注射组(SHED-CA+SHED,n=8)仅去除T8、T9椎板组(SHAM,n=6),建模不治疗组(SCI,n=6)。观察12周分别从大鼠后肢行为学恢复、组织学及神经营养因子分泌等方面进行疗效比较。结果:BBB评分显示SHED-CA+SHED组高于SHED-CA组;免疫荧光染色结果显示SHED-CA+SHED组脊髓损伤区瘢痕组织更少,且再生出更多的神经纤维及髓鞘细胞,并出现SHED向髓鞘细胞的分化;ELISA检测结果提示,联合SHED悬液注射可分泌更多的神经营养因子。结论:联合SHED悬液注射可提高SHED聚合体治疗大鼠脊髓损伤的疗效,可能通过增加神经营养因子分泌及SHED定向分化为髓鞘细胞而发挥作用。Objective:To explore the effective therapy for spinal cord injury in rats based on SH ED aggregates.Methods:Complete transectional injury model of spinal cord was established in 28 SD rats,the rats were dividen into 4 groups:SHED aggregates group(SHED-CA,n=8),group of SHED aggregates combined SHED suspension injection(SHED-CA+SHED,n=8),control groups were SHAM(removal of T8 and T9 vertebral lamina,n=6)and SCI(model without treatment,n=6).After 12 weeks of observation,the therapeutic effects were compared by behavioral recovery of the hind limbs(BBB sore),histology and secretion of neurotrophic factors.Results:BBB score of SHED-CA+SHED group was higher than that of the SHED-CA group(P<0.05).The immunofluorescence staining showed that SHED-CA+SHED group had less scar tissue and more regeneration of neurofilaments and myelin cells in the spinal cord injury area,and SHED differentiated into myelin cells.ELISA test suggested that there were more neurotrophic factors secreted in SHED-CA+SHED group.Conclusion:SHED suspension injection can improve the efficacy of SHED aggregates in the treatment of spinal cord injury in rats,which may play a role in increasing the secretion of neurotrophic factors and inducing the differentiation of SHED into myelin cells.
