摘 要:以‘库尔勒香梨’、‘黑酸梨’、‘沙梨01号’为对照,利用流式细胞仪和根尖压片法对‘棉梨’的染色体倍性进行鉴定,以期为梨种质资源的创新和利用提供参考。结果表明:根尖压片得出‘库尔勒香梨’、‘黑酸梨’、‘沙梨01号’以及‘棉梨’的染色体数目分别为34、51、68以及34,‘棉梨’的染色体数目与二倍体的‘库尔勒香梨’相同;利用流式细胞仪得出‘库尔勒香梨’、‘黑酸梨’、‘沙梨01号’以及‘棉梨’均出现了一个峰,且峰值比为1.0∶1.5∶2.0∶1.0,即各梨品种叶片细胞中DNA相对含量为2C∶3C∶4C∶2C。综上可得:‘棉梨’的染色体数目为2n=2X=34,为单纯的二倍体。With pear varieties Korla Fragrant Pear,Heisuanli and Shali 01 as internal reference standards,the ploidy of Mianli(Pyrus sinkiangensis)was identified by flow cytometry and root-tip squashing methods.The objective was to provide references for the innovation and utilization of pear germplasm resources.The results showed that the chromosome numbers of Korla Fragrant Pear,Heisuanli,Shali 01 and Mianli were 34,51,68 and 34,and the chromosome number of Mianli was the same as that of Korla Fragrant Pear.In addition,one peak was obtained in Korla Fragrant Pear,Heisuanli,Shali 01 and Mianli by flow cytometry,and the peak ratio was 1.0∶1.5∶2.0∶1.0,that was,the relative DNA content in the leaf cells of each pear variety was 2C∶3C∶4C∶2C.In conclusion,the chromosome number of Mianli was 2n=2x=34,a simple diploid.
