摘  要:目的探讨肉豆蔻提取物对慢性缺血引起的大鼠脉络丛和脑组织结构损伤的保护作用。方法选取30只雄性Wistar大鼠,随机分为对照组、慢性缺血缺氧模型组、肉豆蔻干预组,运用HE染色技术,检测大鼠脑组织脉络丛及脑组织内结构的变化;运用TUNEL染色技术,检测脉络丛上皮细胞及血管内皮细胞凋亡情况;运用免疫组织化学染色技术,检测大脑皮质小胶质细胞激活情况。结果肉豆蔻提取物明显抑制了慢性缺血缺氧引起的脉络丛上皮细胞和血管内皮细胞的变性和坏死以及小胶质细胞的大量激活。结论肉豆蔻提取物通过抑制脉络丛上皮细胞和血管内皮细胞凋亡和抑制小胶质细胞的过度激活来实现大脑微环境的稳定。Objective The purpose of this study was to investigate the protective effects of nutmeg extract on the structural damage of choroid plexus and brain tissue in chronic hypoxia ischemia rats. Methods Thirty male Wistar rats were randomly divided into the control group, the chronic ischemic hypoxia model group and the nutmeg intervention group. The structural changes of choroid plexus and brain tissue were detected by HE staining, and the apoptosis of choroid plexus epithelial cells and vascular endothelial cells was detected by TUNEL staining. Immunohistochemical staining was used to detect the activation of microglia in cerebral cortex. Results The nutmeg extract significantly inhibited degeneration and necrosis of choroid plexus epithelial cells and vascular endothelial cells in chronic hypoxia ischemia model rats, including the activation of microglia. Conclusion The nutmeg extract can stabilize the microenvironment of brain through inhibiting the apoptosis of choroid plexus epithelial cells and vascular endothelial cells and the overactivation of microglia.
