摘 要:目的探讨人绒毛膜促性腺激素(hCG)注射日高孕酮(P)对卵巢正常反应者种植窗期子宫内膜基因表达变化的影响。方法选取2014年4月至2015年12月在郑州大学第二附属医院生殖医学中心行长方案体外受精-胚胎移植(IVF-ET)助孕卵巢正常反应患者12例,该次取卵周期取消移植。按hCG日P水平分组:高P组6例(P≥7.13nmol/L);P未升高组6例(P<7.13nmol/L)。取卵后5d采集子宫内膜组织,RNA-seq分析两组内膜基因表达变化,筛选差异表达基因,qRT-PCR验证差异基因,Western blot检测差异基因产物蛋白水平表达,分析相关基因功能。结果两组之间共有31个显著性差异基因。qRT-PCR验证,其中SLC7A4、DDX5、IFI6、HLA-DRB4、HLADRB5、SCGB1D2与RNA-seq结果一致。Western blot结果显示,高P组中SLC7A4、IFI6、HLA-DRB4、HLA-DRB5蛋白表达量低于P未升高组,而DDX、SCGB1D2蛋白表达量高于P未升高组。结论hCG日高P可能影响了种植窗期子宫内膜容受性相关基因表达,为进一步探索其具体分子作用机制提供相应基因靶点。Objective To investigate the effects of high progesterone(P)level on hCG day on endometrial gene expression during implantation window in the moderate ovarian responders.Methods The moderate ovarian responders,who underwent long-protocol in vitro fertilization and embryo transfer(IVF-ET)and cancelled implantation of this oocyte retrieval cycle in the Second Affiliated Hospital of Zhengzhou University between April 2014 and December 2015,were enrolled in this study.According to P level on the administration day of human chorionic gonadotrophin(hCG),patients were divided into increased P group(P≥7.13nmol/L,n=6)and non-increased P group(P<7.13nmol/L,n=6).Endometrial tissues were collected 5 days after oocyte retrieval,and the alteration of gene expression between the two groups was analyzed by RNA-seq to screen for differentially expressed genes.Partial differentially expressed genes possibly associated with endometrial receptivity were verified by qRT-PCR,the protein expression level of the differentially expressed gene were analyzed by Western blot and their functions were analyzed.Results A total of 31 significantly differentially expressed genes between the two groups.The qRT-PCR verified that the expressions of SLC7A4,DDX5,IFI6,HLA-DRB4,HLA-DRB5 and SCGB1D2 were consistent with RNA-seq resluts.Western blot results showed that the protein expression levels of SLC7A4,IFI6,HLA-DRB4 and HLA-DRB5 in the increased P group were lower than those in the non-increased group,while the expression levels of DDX and SCGB1D2 were higher.Conclusion High P on hCG day may influence the endometrial receptivity-related genes expression during implantation window,which provides gene targets for further investigation into the underlying molecular mechanisms.
