摘 要:目的探讨不同期别矽肺病人支气管肺灌洗液中miR-146a和IL-1β的表达特征及其相互关系。方法以35名接受支气管肺灌洗治疗的人群为研究对象,采用RT-qPCR法测定研究对象肺灌洗液中miR-146a相对表达量,ELISA法测定IL-1β表达水平。采用t检验、方差分析等方法分析其表达谱特征及其相互关系。结果不同期别矽肺病人肺灌洗液miR-146a及IL-1β表达水平均呈现先上升后下降的趋势,其中,miR-146a相对表达量在贰期达到最高水平(P<0.05),IL-1β表达量在壹期达最高水平(P<0.05),并且壹期、贰期矽肺患者miR-146a及IL-1β的表达水平均高于对照组(P<0.05);肺灌洗液miR-146a相对表达量与IL-1β表达水平的相关系数为0.457(P<0.05)。结论不同期别矽肺病人肺灌洗液中miR-146a与IL-1β的表达水平不同,二者之间存在相关性,提示miR-146a主要在矽肺早期发挥调控作用,并且表达水平与初次接尘年龄、工龄、饮酒等有关。Objective To discuss the expression characteristics and correlation of miR-146a and IL-1βin silicosis patients of different stages.Methods 35 volunteers experienced bronchoalveolar lavage therapy were enrolled.And then,the expression level of miR-146a and IL-1βin macrophages of bronchoalveolar lavage fluid were determined by RT-qPCR and ELISA,respectively.Analysis of variance,t-test and other statistical methods were used to study the expression profiling of miR-146a and IL-1βand their correlation.Results The expression levels of miR-146a and IL-1βin macrophage in lavage fluid of silicosis patients in different stages were different,but both of which showed a rising first and then declining trend.The relative expression levels of miR-146a and IL-1βreached the highest level in stage II and stage I respectively,with statistically significant differences(P<0.05),and both of them in stage I and II silicosis patients were higher than those in control group(P<0.05).There was a positive correlation(r=0.457)between the expression levels of miR-146a and IL-1β(P<0.05).Conclusion The expression levels of miR-146a and IL-1βin lavage fluid of silicosis patients in different stages are different.At the same time,the expression level of IL-1βis correlated with the expression of miR-146a.Result indicates that miR-146a plays a regulatory role in the early stage of silicosis and its expression level is related to the initial age of dust exposure,working years,drinking,etc.
