摘 要:目的探讨木犀草素对哮喘幼鼠肺组织及血清诱导型一氧化氮合酶(iNOS)及一氧化氮(NO)的影响。方法36只BALB/c幼鼠,雌雄不限,随机分为对照组、哮喘组和木犀草素组,卵蛋白致敏的方法制备哮喘幼鼠模型。收集支气管肺泡灌洗液(BALF),进行细胞总数计数及嗜酸性粒细胞计数,免疫组织化学方法与Western blot方法检测各组幼鼠肺组织iNOS蛋白表达,硝酸还原酶法检测肺组织iNOS活性及NO含量,ELISA方法检测肺组织白细胞介素-4(IL-4)及干扰素-γ(IFN-γ)水平。结果细胞计数结果显示,木犀草素能够降低哮喘幼鼠BALF细胞总数及嗜酸性粒细胞数(P<0.01)。免疫组织化学与Western blot结果显示,木犀草素能够降低哮喘幼鼠肺组织iNOS蛋白表达(P<0.01)。硝酸还原酶法检测结果显示,木犀草素能够降低哮喘幼鼠肺组织iNOS活性及NO含量(P<0.01)。ELISA结果显示,木犀草素能够降低哮喘幼鼠肺组织IL-4表达水平,上调哮喘幼鼠肺组织IFN-γ表达水平(P<0.01)。结论木犀草素能够降低哮喘幼鼠气道炎症,其机制可能与抑制诱导型一氧化氮合酶/一氧化氮信号途径有关。Objective To investigate the effects of luteolin on inducible nitric oxide synthase(iNOS)and nitric oxide(NO)in lung tissue and serum of asthmatic young mice.Methods 36 BALB/c mice,male and female,were randomly divided into control group,asthma group and luteolin group.The asthma model was established by ovalbumin sensitization.Bronchoalveolar lavage fluid(BALF)was collected to count the total number of cells and eosinophils.The expression of iNOS protein was detected by immunohistochemistry and Western blot.The activity of iNOS and the content of NO in lung tissue were detected by nitrate reductase method.The level of iterleukin-4(IL-4)and interference-γ(FN-γ)in lung tissue were detected by ELISA.Results Luteolin could decrease the total number of BALF cells and eosinophils in asthmatic young mice(P<0.01).Immunohistochemical and Western blot results showed that luteolin could reduce the expression of iNOS protein in lung tissue of asthmatic young mice(P<0.01).The results of nitrate reductase assay showed that luteolin could decrease iNOS activity and NO content in lung tissue of asthmatic young mice(P<0.01).ELISA results showed that luteolin could decrease the expression of IL-4 in lung tissue of asthmatic young mice and increase the expression of IFN-gamma in lung tissue of asthmatic young mice(P<0.01).Conclusion Luteolin can reduce airway inflammation in asthmatic young mice,and its mechanism may be related to the inhibition of inducible nitric oxide synthase/nitric oxide signaling pathway.
