摘  要:在体外培养人皮肤瘢痕组织成纤维细胞时,用长春新碱(VCR)刺激后,采用TUNEL染色观察成纤维细胞的凋亡情况;用流式细胞技术测定成纤维细胞的凋亡率;Western blot检测细胞凋亡相关蛋白、内质网应激通路相关蛋白的变化,探讨VCR通过内质网应激途径诱导成纤维细胞凋亡。结果表明:细胞经VCR刺激后, TUNEL染色和流式细胞技术检测结果提示在VCR刺激后成纤维细胞明显发生凋亡。Western blot检测显示经VCR处理后成纤维细胞中促凋亡蛋白表达升高,内质网应激标记蛋白和内质网应激信号通道蛋白的表达上升;而内质网通道抑制剂4-苯基丁酸钠盐可有效逆转这一现象。这一研究提示VCR可诱导成纤维细胞凋亡,而这一现象可能是通过激活内质网应激途径而实现的。Human fibroblasts from skin scar tissue were cultured in vitro and treated with vincristine(VCR). TUNEL staining was used to observe the apoptosis of fibroblasts;flow cytometry was used to detect the apoptosis rate;Western blot was used to detect the changes of apoptosis related proteins, endoplasmic reticulum stress related proteins. After VCR treatment, the results of TUNEL staining and flow cytometry showed that VCR could induce fibroblasts apoptosis. The effect and mechanism of VCR on inducing fibroblasts apoptosis via activating endoplasmic reticulum stress pathway were investigated. Western blot showed that the expressions of apoptotic protein, endoplasmic reticulum stress marker protein and endoplasmic reticulum stress signal channel protein increased after VCR treatment, while the inhibitor of endoplasmic channel(4-phenyl butyric acid, 4-PBA) can effectively reverse this phenomenon. In conclusion, VCR could induce fibroblasts apoptosis, which may be achieved by activating endoplasmic reticulum stress pathway.
