摘  要:目的探究NFAT5基因DNA甲基化程度与激素抵抗型哮喘是否存在关联。方法以20例确诊激素抵抗型哮喘患者为激素抵抗型哮喘组,20名普通哮喘患者为普通哮喘组,20例健康者为正常对照组。检测各组NFAT5基因DNA甲基化水平,比较三组间DNA甲基化程度是否存在差异。结果检测NFAT5基因检测片段的CpG位点总数30个,实际检测到的CpG位点数22个,依次为CpG1、CpG5.6.7、CpG8.9.10.11、CpG12.13、CpG14、CpG15.16、CpG17、CpG18.19、CpG20.21.22、CpG23、CpG24.25;各组间NFAT5基因片段平均DNA甲基化水平差异均无统计学意义(P> 0.05),三组间两两比较组间各个CpG位点甲基化水平,其差异均无统计学意义(P值均> 0.017)。结论本研究暂未发现NFAT5基因在激素抵抗型哮喘、普通哮喘患者中有异常甲基化,NFAT5基因甲基化是否参与激素抵抗型哮喘的发病尚需进一步研究。Objective To explore the association between DNA methylation of NFAT5 gene and hormone resistant asthma.Methods 20 patients with confirmed hormone resistant asthma were selected as the hormone resistant asthma group, 20 patients with common asthma were selected as the common asthma group, and 20 healthy patients were selected as the normal control group. The DNA methylation level of NFAT5 gene in each group was detected, and the difference of DNA methylation among the three groups was compared. Results There were a total of 30 CpG sites detected in NFAT5 gene detection fragments, and 22 CpG sites were actually detected, namely CpG1, cpg5.6.7, cpg8.9.10.11, cpg12.13, CpG14, cpg15.16, CpG17, cpg18.19, cpg20.21.22, CpG23 and cpg24.25. There was no statistically significant difference in the mean DNA methylation level of NFAT5 gene fragment between each group(P > 0.05), and there was no statistically significant difference in the methylation level of each CpG site in pairwise comparison between the three groups(P = > 0.017). Conclusion In this study, abnormal methylation of NFAT5 gene has not been found in patients with hormone-resistant asthma and common asthma. Whether methylation of NFAT5 gene is involved in the pathogenesis of hormone-resistant asthma, it still needs further study.
