摘 要:目的探讨与分析实时荧光定量PCR仪用于流感病毒中的应用效果。方法搜集2018年10月至2019年3月期间对我市哨点医院在流感流行期疑似流感病毒样本共530例作为检测对象,依据检查方案不同分配成参照组(细胞培养法)及实验组(实时荧光定量PCR法),统计两组检测方法阳性检出率及实施荧光定量PCR灵敏度及特异性情况,并予以分析和总结。结果通过对530例疑似流感病例中实验组检测出阳性样本共89份,其中检出新甲H1型71份、H3亚型14份、B型4份;参照组分离毒株57株,其中新甲H1型52株、H3亚型3株,B型2株,各组均未检出H5、H7、H9亚型。则研究组与参照组呈正相关,且新甲H型、H3型、B型与毒株分别呈正相关,P<0.05;实验组流感病毒阳性率为89(16.79)%,参照组流感病毒阳性率为57(10.75)%,差距明显(P<0.05)。结论采用实时荧光定量PCR技术可对流感病毒的特异性及灵敏度更高,可作为首选的检测方法。Objective To explore and analyze the application effect of real-time fluorescence quantitative PCR in influenza virus.Methods In the period from October 2018 to March 2019,a total of 530 cases of suspected influenza virus in the epidemic period of the influenza in the city’s sentinel hospital were collected as the detection target,and the reference group(cell culture method)and the experimental group(real-time fluorescence quantitative PCR method)were assigned according to the inspection protocol.The positive rate of the two groups of detection methods and the sensitivity and specificity of the quantitative PCR were analyzed and summarized.Results A total of 89 positive samples were detected in the experimental group of 530 suspected influenza cases,including 71 new methylH1,14 H3 subtypes and 4 type B,and 57 strains isolated in the control group,including 52 new methylH1,3 H3 and 2 type B,none of which were H5,H7 and H9.The positive rate of influenza virus was 89(16.79)%in the experimental group and 57(10.75)%in the control group(P<0.05).The positive rate of influenza virus in the experimental group was 89(16.79)%,and the positive rate of influenza virus in the control group was 57(10.75)%.There was a significant difference between the two groups(P<0.05).Conclusion The real-time fluorescence quantitative PCR technology can be used as a preferred method for detecting the specificity and sensitivity of the influenza virus.
