摘 要:目的:对骨髓间充质干细胞(BMSCs)进行分析,找到为何复合关节软骨脱细胞基质可以对兔关节软骨部进行修补的原因。方法:使用新西兰为实验材料,将BMSCs从体内分离出来,并在实验室内对其进行繁殖。同时准备好关节软骨脱细胞基质。建立关节软骨部分缺损模型。将实验对象随机分为1组、2组、3组和4组,1组将软骨切除后再原位进行回植,2组使BMSCs中软骨脱细胞基质恢复其缺损部分;3组只通过脱细胞软骨基质来恢复缺损;4组为软骨缺损处不做任何处理。在实验后的4周、8周、12周解剖实验动物,用大体、组织切片进行检验,实验结果用Wakitani评分量表进行评估。结果:在进行实验的12周后,1组回植自体软骨均存活,与正常对比无明显差异,2组得到了一种比较透明的软骨样组织,其缺损恢复速度比3组、4组快得多。结论:通过实验可确定BMSCs中软骨脱细胞基质可以快速恢复好兔关节软骨缺损部分。Objective Bone marrow mesenchymal stem cells(BMSCs) were analyzed to find out why the composite acellular matrix could repair the articular cartilage of rabbits.Method Bone marrow mesenchymal stem cells(BMSCs) were isolated from New Zealand and propagated in the laboratory. Meanwhile, the acellular matrix of articular cartilage was prepared. To establish a partial articular cartilage defect model. The subjects were randomly divided into four groups: 1, 2, 3 and 4. group 1 resected the cartilage and replanted it in situ. In the two groups, the acellular matrix of bone marrow mesenchymal stem cells was used to restore the defect;in group 3, the acellular cartilage matrix was used to restore the defect;in group 4, the cartilage defect was not treated. At the 4 th, 8 th and 12 th week after the experiment, the animals were dissected and examined by gross and tissue sections. The results were evaluated by Wakitani scale.Results After 12 weeks of the experiment, the autogenous cartilage of group 1 survived, and there was no significant difference between the two groups. Group 2 obtained a relatively transparent cartilage like tissue, and its defect recovery speed was much faster in group 3 and group 4.Conclusion We know that cartilage acellular matrix in BMSCs can quickly recover the articular cartilage defects of rabbits.
