摘  要:目的:探索微生物发酵三七皂苷抗脑缺血和抗血栓形成的作用。方法:通过复制脑血栓、脑缺血和颈动脉血栓模型,利用Evans染色等多种方法检测微生物发酵三七皂苷对脑血栓、颈动脉血栓、脑缺血、血栓溶解、耳廓微循环、毛细血管通透性的影响。结果:100、200mg/kg的微生物发酵三七皂苷均明显降低脑缺血大鼠的脑水含量,电镜检结果显示100mg/kg及200mg/kg微生物发酵三七皂苷组海马区多数细胞正常,个别细胞固缩或轻度水肿。有髓、无髓神经纤维及神经毡未见明显异常。200 mg/kg微生物发酵三七皂苷还显著延长小鼠断头后的喘气时间,作用强于等剂量的血塞通胶囊。与生理盐水模型组比较,100、200 mg/kg微生物发酵三七皂苷及血塞通胶囊组的右脑Evans吸光度与右脑重之比值均显著降低(*P<0.05,**P<0.01);100mg/kg、200 mg/kg微生物发酵三七皂苷可明显增大Evans在脑组织中的含量,呈剂量依赖性明显增加小鼠耳廓细动脉、细静脉管径及增多耳廓毛细血管数量,抑制毛细血管通透性。100mg/kg和200 mg/kg的微生物发酵三七皂苷提高栓塞颈动脉的再通数,明显降低颈动脉持续栓塞率,同时显著增加血管的持续再通率,且200 mg/kg微生物发酵三七皂苷的溶栓作用与等剂量的血塞通胶囊相当。结论:微生物发酵三七皂苷具有抗脑缺血和抗血栓形成的作用,其机制可能与提高耐缺氧能力,减轻脑水肿和神经细胞的坏死,改善微循环及溶栓作用有关。Objective:To investigate the anti-brain ischemia and antithrombotic effect of Panax notoginseng from microorganism.Methods:After replicating models of cerebral thrombosis and carotid thrombosis,the effects of microorganism of Panax notoginseng on cerebral thrombosis,thrombolysis,auricle microcirculation,capillary permeability were detected by Evans blue staining and immunoglobulin dissolution methods,and so on.Results:200 mg/kg Panax notoginseng from microorganism significantly decreased the brain water content in rats with cerebral ischemia.Electroscopy showed that most cells in the hippocampus were normal,individual cells were fixed or slightly edematous after 100 mg/kg and 200 mg/kg Panax notoginseng treatment.There was no obvious abnormality in myelinated,unmyelinated nerve fibers and felt.200 mg/kg Panax notoginsen saponins also significantly prolonged the wheezing time of mice after decapitation,and the effect was stronger than that of Xuesaitong capsule with equal dose.compared with the normal saline and cerebral thrombosis model groups,100 and 200 mg/kg of Panax notoginseng and 200 mg/kg Xuesaitong capsule markedly decreased the ratio of right brain Evans blue absorbance to right brain weight;the thrombolytic effect of Panax notoginseng from 200 mg/kg was comparable to the same dose of Xuesaitong capsule.100 and 200 mg/kg of Panax notoginseng significantly increased the content of Evans in brain tissue,the quantity of auricle arterioles,vein diameters and auricle capillaries of mice were increased and capillary permeability was suppressed in dosedependent manner.Meanwhile,100 and 200 mg/kg of Panax notoginseng increased the recanalization number of embolized carotid artery and increased the rate of continuous recanalization of blood vessels,the rate of continuous carotid artery embolization was significantly decreased.Conclusion:The Panax notoginseng from microorganism has anti-brain ischemia and antithrombotic effect,and its mechanism may be related to improving the ability of hypoxia tolerance,reducing
