基于网络药理学探讨肾病Ⅱ号方君药药对治疗IgA肾病血尿的作用机制

摘　　要：目的基于网络药理学探讨肾病Ⅱ号方君药“熟地黄-白茅根”药对的主要活性分子、靶点及治疗IgA肾病血尿的作用机制。方法首先根据口服生物利用度和类药性,在TCMSP中寻找与熟地黄、白茅根相关的活性分子,并利用TCMSP和BATMAN-TCM寻找其靶点;运用GeneCards数据库,搜索IgA肾病血尿相关靶点。采用Cytoscape软件构建活性分子-靶点网络,将上述获取的两类靶点信息利用STRING平台进行GO富集分析,并且分别利用STRING、ClueGO和DAVID数据库进行KEGG信号通路分析。结果从“熟地黄-白茅根”中筛选出的18个活性分子,作用于70个靶点,其中与IgA肾病血尿相关的靶点有27个;所涉及的通路中,显著性高的有VEGF信号通路、利什曼病、钙信号通路。结论初步探讨肾病Ⅱ号方君药“熟地黄-白茅根”的配伍合理性及其治疗IgA肾病血尿的科学性,并为进一步深入研究该药对治疗IgA肾病血尿的作用机制奠定理论基础。Objective To explore the main active molecules,targets of monarch drug(Rehmanniae Radix Praeparata and Imperatae Rhizoma)of Shenbing Decoction Ⅱ and the mechanism of“Rehmanniae Radix Praeparata-Imperatae Rhizoma”in the treatment of IgA nephropathy(IgAN)hematuria based on network pharmacology.Methods Firstly,combined with oral bioavailability and drug-like properties,the active molecules associated with Rehmanniae Radix Praeparata and Imperatae Rhizoma were searched in the TCMSP,and search for targets in the TCMSP and BATMAN-TCM.Search for IgAN hematuria-related targets in the GeneCards database.Then,the active molecular-target network was constructed by Cytoscape software.The acquired target information using STRING platform for GO enrichment analysis,and using STRING,ClueGO and DAVID databases for KEGG signaling pathway analysis.Results Eighteen active molecules were screened from“Rehmanniae Radix Praeparata-Imperatae Rhizoma”,which acted on 70 targets,and 27 targets related to IgAN hematuria.The highly significant pathways were VEGF signaling pathway,Leishmaniasis,Calcium signaling pathway,etc.Conclusion The rationality of the compatibility of monarch drug(Rehmanniae Radix Praeparata and Imperatae Rhizoma)of Shenbing Decoction Ⅱ and its scientific of IgAN hematuria treatment were preliminarily verified,and the theoretical basis for further study on the mechanism of treatment of IgAN hematuria with these two drugs was established.
