乐孕宁口服液对妊娠缺血性贫血大鼠的作用机制研究

摘　　要：目的:基于多项评价指标系统研究乐孕宁口服液对妊娠期缺铁性贫血的改善作用,并初步探索其作用机制。方法:采用低铁饲料干预联合定期放血的方式建立大鼠缺铁性贫血模型,将造模成功的大鼠与雄鼠按照2:1比例合笼,使其受孕。观测不同时期大鼠的形态和外观变化,并称定不同阶段大鼠的体重。采用全自动血球分析仪检测不同时期大鼠血液的血红蛋白(Hb)、红细胞(RBC)、平均红细胞体积(MCV),红细胞压积(Hct),红细胞体积分布宽度(RDW)等各项血液学指标;测定不同分组大鼠的心肌和肝脏指数;采用ELISA法测定FEP、血清铁和组织中铁的含量,并测定血清中Fn的含量;免疫组化法检测心肌Tfr1和Tfr2表达。结果:乐孕宁口服液不同剂量给药组均可不同程度改善妊娠IDA引起的体重增长迟缓,对心肌和肝脏指数有一定的影响;可显著升高Hb、RBC、Hct、MCV指标,降低RDW;妊娠IDA给予不同剂量乐孕宁口服液后,血清和组织中铁的含量均有不同程度的升高,且血清中Fn含量也有升高趋势;免疫组化测定结果显示,给药组心肌组织中TfR1和Tfr2水平相较于Model组有一定程度的升高。结论:乐孕宁口服液可在一定程度上改善妊娠铁缺乏,若与补铁剂联合使用效果更佳,可达到事半功倍的效果。Objective Based on a number of evaluation indicators,this study systematically explored the effect of Leyunning oral liquid on iron deficiency anemia during pregnancy,and preliminarily explained its mechanism.Metliods The rat model of iron deficiency anemia was established by low iron diet intervention combined with regular bloodletting,and the morphology and hematological indexes of rats in different periods were observed.Results After drug intervention,the hematological parameters and the expression of ferritin in pregnant IDA rats could be improved to a certain extent.Leyunning oral liquid showed a dose-dependent effect in the treatment of pregnancy IDA.After high-dose drug intervention,there was no statistical difference in many indexes between Leyunning oral liquid and control group.Of all the drug groups.Leyunning oral liquid combined with iron supplement showed a better effect after dry intervention.Conclusion Leyunning oral liquid can improve pregnancy iron deficiency to a certain extent.If combined with iron supplement,the effect is better,and the effect of twice the result with half the effort can be achieved.
