摘 要:目的:建立食蟹猴CCl4急性肝损伤模型并观察藤茶总黄酮对食蟹猴CCl4急性肝损伤的保护作用。方法:对食蟹猴腹腔注射1.04%、0.52%、0.26%的CCl4花生油溶液,注射体积2 mL/kg,检测各浓度不同时间点食蟹猴血清转氨酶含量变化,确定有效建立CCl4所致食蟹猴急性肝损伤浓度及时间;对食蟹猴灌胃给予藤茶总黄酮高剂量(128 mg/kg)、低剂量(64 mg/kg),连续10 d,建立食蟹猴CCl4急性肝损伤模型,检测用药后急性肝损伤食蟹猴血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)的含量,并对肝组织进行病理切片及检查。结果:以注射体积2 mL/kg腹腔注射1.04%CCl4花生油溶液后,在16 h时间点血清ALT及AST含量最高(P<0.05);藤茶总黄酮高、低剂量组与模型组比较,细胞水肿、脂肪变性及坏死程度明显减轻,并显著降低急性肝损伤食蟹猴血清ALT的含量(P<0.05);藤茶总黄酮高剂量组可显著降低急性肝损伤食蟹猴血清AST的含量(P<0.05)。结论:腹腔注射2 mL/kg的1.04%浓度CCl4花生油溶液16 h,可成功建立食蟹猴急性肝损伤模型;藤茶总黄酮对CCl4致食蟹猴急性肝损伤有保护作用。Objective:To establish a model of acute liver injury induced by CCl4 in Macaca fascicularis and to observe the protective effect of total flavone of Ampelopsis Grossedentata on this model.Methods:The Macaca fascicularis were intraperitoneally injected with 1.04%,0.52%,and 0.26%CCl4 peanut oil solution,and the injection volume was 2 mL/kg.The changes of serum transaminase content in Macaca fascicularis at each concentration and at different times were detected,and the effective establishment of the concentration and time of CCl4 induced acute liver injury in Macaca fascicularis were determined.Macaca fascicularis were given with high(128 mg/kg)and low-dose(64 mg/kg)of total flavone of Ampelopsis Grossedentata by intragastric administration for 10 days.Then each group of Macaca fascicularis that treated with total flavone of Ampelopsis Grossedentata were given CCl4 to establish acute liver injury model.The contents of Alanine Aminotransferase(ALT)and Aspartate Aminotransferase(AST)in serum of model Macaca fascicularis were detected,and the pathological sections and examinations of liver tissue were performed.Results:After intraperitoneal injection of 1.04%CCl4 peanut oil solution with an injection volume of 2 mL/kg,the contents of serum ALT and AST were the highest at 16 hthan other time point and concentration(P<0.05).The degree of cell edema,steatosis and necrosis in the high and low dose groups were significantly less than those in the model group,and the content of serum ALT in model Macaca fascicularis was significantly decreased(P<0.05).The high dose of total flavone of Ampelopsis Grossedentata could significantly reduce the content of serum AST in model Macaca fascicularis(P<0.05).Conclusion:The model of acute liver injury in Macaca fasciculari can be successfully established by intraperitoneal injection of 1.04%CCl4 peanut oil solution with an injection volume of 2 mL/kg for16 hours.The total flavone of Ampelopsis Grossedentata has protective effects on acute liver injury induced by CCl4 in Macaca fascicula
