摘 要:目的:探究大黄蟅虫丸对肝炎后肝纤维化大鼠抑制作用的机制。方法:选取60只雄性Wistar大鼠,随机分对照组、模型组及治疗组,每组20只。模型组和治疗组大鼠使用CCl4建立肝纤维化模型,且治疗组灌胃给予1.6 g/kg大黄蟅虫丸,1次/d;对照组、模型组予以等剂量生理盐水,共6周。检测并比较各组大鼠血清中丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、总胆红素(TBil)、清(白)蛋白(Alb)水平;各组体重、肝重量、肝脏指数、纤维化情况;测定肝组织谷氨酰转肽酶(γ-GT)、丙二醛(MDA)、羟脯氨酸(Hyp)、还原型谷胱甘肽(GSH)和超氧化物歧化酶(SOD)活性水平;并通过苏木精-伊红(HE)染色观察肝脏病理情况。结果:模型组与治疗组大鼠的体重低于对照组,肝重量和肝脏指数高于对照组,且治疗组治疗结束时的体重高于模型组,肝重量和肝脏指数均低于模型组(P<0.05)。模型组与治疗组大鼠的血清ALT、AST、TBil水平均高于对照组,Alb水平低于对照组,且治疗组的血清ALT、AST均低于模型组(P<0.05)。模型组与治疗组大鼠的肝组织γ-GT、MDA、Hyp活性均高于对照组,且治疗组均低于模型组;模型组与治疗组大鼠的肝组织GSH和SOD活性低于对照组,且治疗组均高于模型组(P<0.05)。模型组与治疗组大鼠的肝组织透明质酸酶(HA)、层黏连蛋白(LN)、肝纤维化指数水平均高于对照组,且治疗组均低于模型组(P<0.05)。对照组大鼠无成纤维细胞增生,模型组大鼠的肝细胞纤维化情况较治疗组更加严重,多为"++"和"+++"。结论:大黄蟅虫丸可抑制大鼠肝炎后肝纤维化,其作用机制可能与抗肝脂质过氧化反应有关。Objective:To explore the inhibitory effect of Dahuang Zhechong Pill on hepatic fibrosis after hepatitis in rats.Methods:Sixty male wistar rats were randomly divided into control group,model group,and treatment group,with 20 rats in each group.The rats in the model group and the treatment group were treated with CCl4 to establish the model of hepatic fibrosis,and the treatment group was given 1.6 g/kg Dahuang Zhechong Pill once a day,while the control group and model group were given the same dose of normal saline,thewhole experiment would last 6 weeks.The levels of serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),total bilirubin(TBil),albumin(Alb),body weight,liver weight,liver index and fibrosis were detected and compared.The activities of glutamyl transpeptidase(γ-GT),malondialdehyde(MDA),hydroxyproline(Hyp),reduced glutathione(GSH)and superoxide dismutase(SOD)in liver tissue were measured,and the liver pathology was observed by HE staining.Results:In the model group and the treatment group,the body weight of the rats was lower than that in the control group,and the liver weight and liver index were higher than those in the control group.In the treatment group,the body weight at the end of treatmentwas higher than that in the model group,and the liver weight and liver index were lower than that in the model group(P<0.05).In the model group and the treatment group,the serum levels of ALT,AST and TBi were higher than those in the control group,while the level of Alb was lower than that in the control group.In the treatment group,the serum ALT and AST were lower than those in the model group(P<0.05).The activities ofγ-GT,MDA and Hyp in rats’liver tissue in model group and treatment group were higher than those in control group,and thoseindex in treatment group were lower than those in model group.The activities of GSH and SOD in rats’liver tissue in model group and treatment group were lower than those in control group,and those indexin treatment group were higher than those in model group
