摘  要:目的制备携hVEGF165表达载体阳离子脂质微泡并测定其与hVEGF165表达载体的结合率。方法制备携hVEGF165表达载体阳离子脂质微泡,测定其粒径大小、浓度、Zeta电位;测定与hVEGF165表达载体的结合率。结果阳离子脂质微泡90%粒径范围:1μm-3μm,平均粒径:(1.31±0.23)μm,平均浓度:(3.17±0.16)×108/mL,Zeta电位:(28.00±1.84)mV,与hVEGF165表达载体的最大结合率:(35.34±0.71)%。结论采用机械振荡法制备的阳离子脂质微泡,粒径分布良好,浓度、Zeta电位稳定,与hVEGF165表达载体结合率较高,是携带基因表达的良好载体系统。Objective To prepare cationic lipid microvesicles carrying hVEGF165 expression vector and determine its binding rate to hVEGF165 expression vector.Methods Cationic lipid microbubbles containing hVEGF165 expression vector were prepared and their particle size,concentration and Zeta potential were measured.The binding rate to hVEGF165 expression vector was determined.Results The particle size range of 90%cationic lipid microbubbles was 1μm-3μm,the average particle size was(1.31±0.23)μm,the average concentration was(3.17±0.16)×108/mL,the Zeta potential was(28.00±1.84)mV,and the maximum binding rate to hVEGF165 expression vector was(35.34±0.71)%.Conclusion The cationic lipid microbubbles prepared by mechanical oscillation method have good particle size distribution,stable concentration and Zeta potential,and high binding rate with hVEGF165 expression vector.
