摘 要:目的:探讨3D打印羟基磷灰石晶体-可溶性胶原(hydroxyapatite-geltin,HAP-GEL)支架复合骨髓间充质干细胞(bone regeneration bone marrow mesenchymal stem cells,BMSCs)和人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVECs)组织工程骨对兔颅骨缺损的修复效果。方法:将3D打印的HAP-GEL复合物支架与BMSCs和HUVECs共培养;制作颅骨缺损模型,随机分为3组,即单独HAP-GEL支架组,HAP-GEL支架复合细胞组和空白对照组。术后12周对标本进行大体观察和X线检测,分析骨修复情况。结果:大体标本观察,空白对照组仅有少量骨质形成;HAP-GEL组材料有部分降解,骨缺损边缘较清晰;HAP-GEL复合细胞组骨痂形成较多,骨缺损界限不清。X射线显示,空白对照组骨缺损为较明显的透射影;与HAP-GEL组相比,HAP-GEL复合细胞组缺损区域密度明显增高,接近正常骨组织。结论:负载有细胞的HAP-GEL复合体对兔颅骨缺损的修复效果优于单纯HAP-GEL支架材料,是一种相对理想的骨缺损修复替代材料。Objective:To investigate the bone defect repair ability of hydroxyapatite-geltin(HAP-GEL)scaffold with bone marrow mesenchymal stem cells(BMSCs)and human umbilical vein endothelial cells(HUVECs)by in rabbit skull defect.Methods:The 3 D printed HAP/GEL composite scaffold was co-cultured with bone regeneration BMSCs and HUVECs.Make skull defect models and were randomly divided into three groups,which were implanted with HAP-GEL scaffold,HAP-GEL composite BMSCs and HUVECs scaffold,and a blank control group was set up.At 12 weeks after the operation,the specimens were observed by X-ray,and the bone repair was analyzed.Results:From gress inspection,there was only a small amount of bone formation in control group.In alone scaffold group,the scaffold material was partially degraded and the edge between the material and bone tissue was clarityed.The BMSCs+HUVECs+HAP-GEL group had more bone callus formation and unclear bone defect boundary.From X-ray,the bone defect area of the control group was more obvious.Compared with the alone HAP-GEL group,the HAP-GEL loaded with BMSCs and HUVECs cells group showed significant density,which was close to normal bone tissue.Conclusion:Cells combined with HAP-GEL scaffold has a better repair effect on rabbit skull defect than simple HAP-GEL scaffold,which is a relatively ideal substitute for bone defect repair.
